摘要 目的:探讨葎草中生物碱的提取方法及含量测定。方法:采用正交实验方法,以提取温度、提取时间、料液比、盐酸浓度为影响因素,以生物碱得率为指标,优选生物碱最佳提取条件;采用生物碱颜色反应和薄层色谱进行鉴定;采用酸碱滴定方法进行生物碱含量测定。结果:葎草生物碱中含有乌头碱。葎草中生物碱的提取温度70℃、时间60 min、料液比1∶30、盐酸浓度2%为葎草中生物碱的最佳提取条件。精密度实验RSD为2.11%,加样回收试验RSD为1.46%,稳定性试验RSD为1.80%。结论:该工艺条件下葎草中生物碱的提取方法简便、快捷,生物碱得率达0.283%;精密度较高、稳定性较好、方法可行。 Objective: To discusses the extracting process and content determination of alkaloids from Humulus Scandens.Methods: Using alkaloids yield as the index, the extracting process for alkaloid was obtained by orthogonal experiment with extraction temperature, extraction time, solid-liquid ratio, and concentration of hydrochloric acid as the experimental factors. Using alkaloids color identification reaction and TLC identification to verify alkaloids. Acid-base titration was used to measure it. Results:Alkaloids in Humulus Scandens was mainly Aconitine. The best extraction conditions: the temperature of 70 ℃, time for 60 minutes, solid-liquid ratio 1∶30, and hydrochloric acid concentration of 2% for the extraction of alkaloid from Humulus Scandens.RSD was 2.11%, 1.46% and 1.80% in the precision test, the recovery test and the stability test, respectively. Conclusion: The extracting process for alkaloids from Humulus Scandens was simple and fast with high precision recovery and stability. The yield of alkaloids was 0.283%.
