鸡眼草总黄酮含量测定及其植株含量动态积累 被引量:1

摘要 目的:建立操作简单、稳定性好的鸡眼草总黄酮含量测定方法,比较鸡眼草不同采收期的植株生物量及总黄酮积累量.方法:以紫外分光光度法考察鸡眼草提取液在三乙胺介质中的紫外可见光谱;测定鸡眼草植株生物量与不同器官的总黄酮含量,考察总黄酮积累的动态变化.结果:在三乙胺介质中,鸡眼草总黄酮和芦丁的最大吸收均红移至400 nm处;芦丁在1.0~25.0 mg/L浓度范围内与吸光度呈良好的线性关系.不同采收期间鸡眼草根、茎、叶的总黄酮含量差异显著;6~8月总黄酮含量均高于9~11月.8月下旬~9月上旬植株生物量较大,植株总黄酮积累量达到峰值.结论:三乙胺方法用于鸡眼草总黄酮含量测定,方法简便、重现性较好,可操作性强.8月下旬~9月上旬适于大量采集鸡眼草用于生产. Objective: To develop a simple and stable method for determining the total flavonoids of K. striata, and to investigate the plant biomass and total flavone accumulation of plants that were picked regularly in different harvest times. Methods: The suitability of TEA UV/Vis spectrophotometric method was studied by comparing the spectra of the total flavonoids in K. striata. The plant biomass of plants and total flavonoids of different organs were tested. Results : Both of the total flavonoids in K. striata and Rutin could produce a red shift of the maximum absorption in the medium of triethylamine. There was a good linear relationship between the concentrations and the absorbance of Rutin in the range of 1.0 ~ 25.0 mg /L. The yield of flavonoids among 3 parts of plant were significantly different. The yield during June to August was higher than that during September to November. However, in late August and early September, the plant biomass got bigger and the accumulation of total flavonoids of plants reached the maximum value. Conclusions: The TEA method was simple, convenient and reproducible. It could be applied for determining the total flavonoids in Kummerowiaschindl. During late August and early September,it is appropriate to collect the herb in large-scale,which can be used in production.
