摘要
目的:研究重组腺相关病毒(r AAV2/1)介导IGF-1基因对糖尿病ED(DMED)的修复作用及作用机制。方法:建立DMED动物模型,诱导糖尿病8周后,应用阿朴吗啡(APO,80μg/kg)皮下注射进行阴茎勃起试验,筛选DMED大鼠模型。实验分组:A组为正常对照组,B组为模型未治疗组,C组为单纯转染r AAV2/1-IGF-1组,D组为控制血糖组,E组为转染r AAV2/1-IGF-1组+控制血糖组。应用免疫组化法比较IGF-1和NOS基因在各组中的表达水平,检测阴茎海绵体内压。结果:C、D、E组IGF-1和n NOS阳性细胞百分比和IHS评分均高于B组,差异具有统计学意义(P<0.05),E组IGF-1和n NOS阳性细胞百分比和IHS评分分别为(71.90±3.04)%、(4.16±0.15)分和(72.33±7.12)%、(4.32±0.28)分,均高于D组,差异具有统计学意义(P<0.05),C、D、E组阴茎阴茎海绵体内压均高于B组,差异具有统计学意义(P<0.05),E组阴茎阴茎海绵体内压为(230.69±8.34)mm Hg,高于D组,差异具有统计学意义(P<0.05)。结论:应用r AAV2/1介导IGF-1基因对DMED大鼠能够显著提升IGF-1和NOS表达,增加阴茎海绵体内压,对阴茎勃起功能障碍具有显著的修复作用。
Objectives: To study the effect and mechanism of recombinant adenovirus( r AAV2 /1)- mediated IGF- 1 gene in the treatment of diabetic erectile dysfunction( DMED). Methods: The DMED animal model was established. 8 weeks after diabetes induction,penile erection test was done by subcutaneous injection of apomorphine( APO,80μg / kg) and DMED rat model was screened. The group A was control group,group B was untreated group and group C was single r AAV2 /1- IGF- 1 transfected group,group D was blood glucose control group,Group E was r AAV2 /1- IGF- 1 transfected and blood glucose control group. IGF- 1 and NOS expression of each group was detected by immunohistochemical method as well as intracavernous pressure. Results: IGF- 1and n NOS positive cells percentage and IHS scores of group C,D,E were higher than these of group B with statistically significant differences( P〈0. 05). IGF- 1 and n NOS positive cell percentage and IHS scores of group E were( 71. 90 ± 3. 04) %,( 4. 16 ± 0. 15) points and( 72. 33 ± 7. 12) %,( 4. 32 ± 0. 28) points,which were all higher than these of group D with statistically significant differences( P〈0. 05). The penis inside pressure of group C,D,E were higher than that of group B with statistically significant differences( P〈0. 05). Penis inside pressure of group E was( 230. 69 ± 8. 34) mm Hg,which was higher than that of group D with statistically significant difference( P〈0. 05). Conclusion: Application of r AAV2 /1 mediated IGF- 1 gene on DMED rats can significantly improve expression of IGF- 1 and NOS,increase intracavernous pressure,with significant repair effect on erectile dysfunction.
