摘要
人体的肝脏、肾脏和肠道上广泛分布着多种转运蛋白,其中多药及毒物外排转运蛋白(MATEs)主要表达在肾脏近端小管的刷状缘膜及肝细胞胆小管面,其功能主要是调节有机阳离子的外排。该文主要讨论的是MATEs家族的成员MATE1和MATE2-K的分型、结构、遗传多态性以及其功能意义,同时也就MATEs的遗传多态性对铂类化合物及二甲双胍药物代谢动力学以及药效学的影响进行阐述。
The body is equipped with kinds of transporters which generally exist in liver,kidney,and intestine. Multidrug and toxin extrusion proteins( MATEs; SLC47A) are predominantly expressed in the brush-border membrane of proximal tubule epithelial cells in the kidney and the canalicular membrane of hepatocytes. Functionally,MATEs act as efflux transporters for organic compounds. The article discusses type,structure,polymorphism and function of MATE1 and MATE2-K,and also discusses the effect of single nucleotide polymorphisms( SNPs) in the SLC47A1 gene and SLC47A2 on the pharmacokinetics and pharmacodynamics of metformin and platinum-based chemotherapeutic agents.
