摘要
对小鼠原代肝细胞分离方法进行改良,通过简单且经济的方法获得小鼠原代肝细胞,采用4.5号输液针经下腔静脉逆行灌流,用长条状纸板对针进行固定,以获取质量稳定的小鼠原代肝细胞悬液.结果显示,改进后的方法灌流成功率高及重现性好并可获得实验所需肝细胞,细胞对台盼蓝拒染率>90%.方法操作简单,成本较低且效率较高.
The paper studies how to improve the separation method for mouse primary hepatocyte in order to obtain the mouse primary hepatocyte in the most simple and economic way. Retrograde infusion is done by using infusion needles through the inferior vena cava. Then, the needles are fixed by long strip paperboard. In this way, stable suspension of mouse primary hepatocyte can be obtained. As a result, the success rate of the improved retrograde infusion is extremely high and the repetition is also excellent. The hepatocyte needed for experiments can be obtained. The rate of trypan blue stain-resistance is more than 90%. This method is simple in operation, low in cost and high in efficiency.
