17β-estradiol enhances periostin expression in BMSCs of ovariectimized rats

摘要 目的:探讨雌激素对大鼠骨髓间充质干细胞(r BMSCs)中Periostin表达的影响。方法:取6周龄雌性SD大鼠,分为去势大鼠模型(OVX)、假手术(Sham)、OVX+17β-雌二醇(17β-E2)组。取Sham、OVX组r BMSCs进行分离、培养,流式细胞术鉴定细胞表面分子标记物;成骨诱导各组r BMSCs的成骨分化,碱性磷酸酶(ALP)和茜素红染色检测成骨分化情况;RT-PCR检测3组r BMSCs中Periostin、Runx2、OCN、BSP的表达水平。结果:成功构建OVX;与Sham组相比,OVX组ALP及茜素红显色均较浅,OVX+17β-E2组能加深ALP及茜素红染色。RT-PCR结果显示,OVX组Periostin、Runx2、OCN、BSP表达显著降低;OVX+17β-E2组的Periostin、Runx2、OCN、BSP表达水平均明显升高。结论:雌激素可显著提高去势大鼠BMSCs中Periostin的表达。 AIM: To investigate the effects of 17 β-estradiol ( 17β-E2 ) on the expression of periostin in rBMSCs of ovariectimized rats. METHODS: Ovariectomy (OVX) was performed to establish osteoporosis in 6 -w SD rats. rBMSCs were separated and cultured from Sham, OVX, and OVX + 17β-estradiol treated rats, and then cell surface markers were indentified by flow cytometry. After osteoblastic induction, esteoblastic differentiation was evalua- ted by ALP activity and alizarin red staining. The expression level of periestin, Runx2, OCN and BSP was determined by RT - PCR. RFULTS: ALP expression and mineralization nodules were evidently observed in sham group, and significantly decreased in OVX group. 17β- E2 treatment increased the ALP expression and mineralization nodule formation. RT-PCR results showed that the expression of periostin, Rnx2, OCN and BSP decreased significantly in OVX rats. 17β- E2 treatment significantly increased the expression of periestin, Runx2, OCN and BSP ( P 〈0.05). CON- CLUSION: 17 β-estradiol can enhance the expression of Periestin in BMSCs of ovaxiectimized rats.
