摘要
目的:探讨热休克基因相关蛋白在牙髓组织增龄性变化过程中的表达。方法:取雄性SD大鼠30只,其中月龄>10个月为老龄组(n=15),<2个月为年青组(n=15),然后取两组大鼠下颌第一磨牙连续切片及牙髓组织,用免疫组化染色,RT-PCR、Western Blot检测比较两组大鼠牙髓组织中热休克转录因子1(HSF1)、热休克蛋白27(HSP27)的表达水平。结果:HSF1和HSP27均表达于SD大鼠牙髓组织中,且HSF1mRNA及蛋白表达水平随大鼠年龄的增加而升高,而其下游的HSP27表达水平却随之下降。结论:热休克基因相关蛋白可能参与牙髓增龄性变化的过程。
AIM : To investigate the age - related changes of heat shock protein(HSP) expression in dental pulp tissue. METHODS: Male SD rats with the ages of 〉 10 m (n = 15) and 〈 2 m (n = 15) were used in this study. The mandibles with the bilateral first molars were sectioned, fixed and decalcified. The expression of heat shock transcription factor 1 ( HSF1 ) and heat shock protein 27 ( HSP27 ) in the dental pulp tissues were examined by immu- nohistochemical staining, RT-PCR and Western blot. RESULTS: Both HSF1 and HSP27 were expressed in the dental pulp tissues of SD rats. The mRNA and protein expression of HSF1 increased with age. However, the mRNA and pro- tein expression of HSP27 decreased with age. CONCLUSION: Heat shock protein might be involved in the process of dental pulp ageing.
