前列腺癌组织中Survivin和HIF-1α的表达及临床意义

Expression and clinical significance of Survivin and HIF-1α in prostate cancer tissue

摘要 目的通过检测前列腺增生(benign prostatic hyperplasia,BPH)和前列腺癌(prostatic cancer,PCa)组织中生存素(Survivin)和缺氧诱导因子-1α(hypoxia inducod factor-1α,HIF-1α)的表达情况,分析Survivin和HIF-1α表达与前列腺癌组织Gleason评分、肿瘤转移的关系及二者间的相互关系。方法采用免疫组织化学S-P法检测60例PCa和40例BPH组织中Survivin及HIF-1α表达情况,并分析两者与前列腺癌临床病理特征的关系及其两者之间的相关性。计数资料比较采用χ2检验,相关性分析采用Spearman等级相关分析,P<0.05为差异有统计学意义。结果 PCa组织中Survivin及HIF-1α的表达情况分别为81.7%和76.6%,二者与BPH组织相比,差异均有统计学意义(均P<0.05)。PCa组织中Survivin和HIF-1α的表达均与Gleason分级、癌穿透被膜、骨转移及淋巴结转移有关,差异均有统计学意义(均P<0.05)。PCa组织中,Survivin与HIF-1α的阳性表达率呈显著正相关(P<0.05)。结论 Survivin和HIF-1α的阳性率越高,PCa的恶性度越高,浸润越深。Survivin和HIF-1α相互作用共同参与PCa的发生、发展。 Objective To detect the expression of inhibitor of Survivin and hypoxia induced factor-1α(HIF-1α)in prostate carcinoma(PCa) and benign prostate hyperplasia(BPH) tissue, and to analyze the relationship of Survivin and HIF-1α gene expression and Gleason score, clinical stage in prostate cancer, and the relationaship between these two antibodies. Methods The expressions of Survivin and HIF-1α in the tissue of PCa(n=60) and BPH(n=40) were detected by immunohistochemistry S-P method, and the relationaship between these two antibodies and the clinic pathological features of PCa were analyzed, so was the correlation between the expressions of Survivin and HIF-1α. Count data was used by chi square test, correlation analysis was used by Spearman rank correlation analysis,P〈0.05 was considered statistically significant.Results The expressions of Survivin and HIF-1α in PCa were 81.7% and 76.6%, there were significant differences(all P〈0.05). The expressions of Survivin and HIF-1α were closely related to the Gleason score, cancer through the tunica, distant bone metastasis and lymph node metastasis(all P〈0.05). The expressions of Survivin and HIF-1α were positively correlated in PCa(P〈0.05).Conclusion There are positive correlations in the expressions of Survivin and HIF-1α in PCa. Survivin and HIF-1α play important roles in development and progression of PCa.
