摘要
为了明确能量代谢负平衡奶牛血清中成纤维细胞生长因子(FGF-21)浓度的变化规律以及与奶牛能量代谢负平衡特征性指标之间的关系。研究从某千头奶牛场选取选择非妊娠、非泌乳、健康奶牛10头,随机分成2组,每组5头。在实验期间,一组牛饲喂120%维持(1.48 Mcal·kg^(-1))日粮,另一组饲喂30%维持能量(0.37 Mcal·kg^(-1))日粮(NRC标准),每天采集血液样品,检测能量代谢特征性指标,其中,血清理化指标采用全自动血液生化分析仪进行测定,激素指标Elisa试剂盒进行检测。所有检测数据均采用统计学方法进行分析与统计。结果显示:摄入30%维持能量(0.37 Mcal·kg^(-1))日粮的牛血清FGF-21水平同时显著升高;饲喂120%的维持日粮的牛血清FGF-21水平同时显著降低。实验检测指标中血清FGF-21指标与瘦蛋白(LP)为正相关,与血糖(Glu),β羟丁酸(BHBA)为负相关。结论:实验表明FGF-21与能量密切相关并且在奶牛能量代谢平衡状态的调控中发挥作用,为今后深入探究能量负平衡的发生机理和新的防治策略奠定了基础。
To investigate the variation and relationship between serum FGF-21 concentration and characteristic parameters related to energy metabolism in blood in dairy cattle with negative energy balance, 10 non-pregnant,non-lactating dairy cattle was randomly allocated in two groups.The interventions were a controlled-energy diet (0.37 Mcal/kg) and a moderate-energy diet (1.48 Meal/kg) during duration of experiment.Biochemical parameters, serum FGF-21 ,leptin and insulin levels were measured by commercial ELISA kits.The results Showed that serum FGF-21 levels were significantly higher in the group treated with a controlled-energy diet,while FGF-21 levels in moderate-energy diet group were the opposite.In a synthesis analysis of two groups,FGF-21 levels were significantly positively correlated with serum leptin levels while an inverse relationship of FGF-21 with blood glucose and BHBA levels was found.An inverse relationship of FGF-21 with BHBA was found.The increase in FGF-21 levels after a controlled-energy diet treatment may contribute to positive metabolic effect and it would be a new theoretical and practical basis on the preventive strategy of dairy cattle with NEB.
