摘要
目的探讨马铃薯提取物对人卵巢癌细胞(HO-8910)体外增殖的影响和可能的作用机制。方法用MTT法检测不同作用时间(24、48、72h)、不同体积比(13、25、50、100μL·mL-1)马铃薯提取物对HO-8910体外增殖的影响,倒置显微镜观察不同马铃薯提取物对HO-8910的形态学改变。应用Western blot技术,在50μL·mL-1马铃薯提取物作用HO-8910细胞后0、20、40、60min Bcl-2和Bax的的表达量。结果作用48h时,50μL·mL-1体积比马铃薯提取物对HO-8910抑制率为(88.44±4.56)%,显著高于其他体积比(P<0.05);马铃薯提取物使Bax表达量均较空白增加,且随时间的增加表达量有上升趋势,Bcl-2蛋白则与之相反,促进细胞凋亡。结论马铃薯提取物对HO-8910增殖具有显著的抑制作用。
Objective To explore the effect of potato extract on proliferation of human ovarian cancer HO-8910 cells and its mechanism of action. Methods The proliferation of HO-8910 cells was detected by MTT after treatment with different concentrations of potato extract(13,25,50, 100 μL·mL^-1) for 24,48 and 72 hours. Morphological changes in HO-8910 cells were observed under an inverted microscope. The expression of Bcl-2 and Bax in HO-8910 cells was measured by Western blot after treatment with 50μL ~ mL^-1 potato extract for 0, 20, 40 and 60 minutes. Results After treatment for 48 hours, the inhibition rate in 50μL·mL^-1 treatment group was significantly higher than that in other groups(P〈0.05). Western blot showed that potato extract increased Bax expression and decreased Bcl-2 expression in a time-dependent manner. Conclusion Potato extract can inhibit the proliferation of HO-8910 cells.
