摘要
目的:探讨盐酸普鲁卡因对体外培养的人白血病HL-60细胞ID4基因甲基化状态的影响。方法:体外培养HL-60细胞,使用不同浓度(2 mmol/L、4 mmol/L、6 mmol/L)的普鲁卡因处理细胞,分别采用MS-HRM、RT-PCR法检测HL-60细胞ID4基因启动子区域的甲基化状态和ID4m RNA的表达。结果:HL-60细胞ID4基因在普鲁卡因作用下呈现明显的去甲基化状态,ID4m RNA表达明显增加,且以上改变均随药物浓度的增加而增强(P<0.05)。结论:盐酸普鲁卡因对HL-60细胞的ID4基因启动子区域有去甲基化作用,上调细胞ID4 m RNA的表达。
Objective: To investigate the effects of procaine hydrochloride on methylation of ID4 gene of human leukemia HL-60 cells. Methods: HL-60 cells cultured in vitro were treated with different concentration (2 mmol/L, 4 mmol/L, 6 mmol/L) procaine hydrochloride, MS-HRM was used to detect the methylation status of the promoter region of ID4 gene in HL-60 cells, the expression of ID4 mRNA and protein was analyzed by RT- PCR. Results: ID4 gene of HL-60 cells was obvious demethylation under the action of procaine, the expression of ID4 mRNA and protein was significantly increased, and the above changes were enhanced with the increase of the concentration of the drug (P〈 0.05). Conclusion: Procaine hydrochloride could decrease the methylation of HL-60 cells in ID4 gene promoter region and upregulating the expression of ID4 gene
