摘要
目的:建立A2780-cp卵巢癌细胞株的NOD scid小鼠原位成瘤模型,为卵巢癌的进一步研究提供参考载体.方法:体外培养A2780-cp卵巢癌细胞,制成细胞密度为5×10^6/mL的单细胞悬液,原位种植:取20μL注入小鼠卵巢皮下;皮下种植:癌细胞悬液0.1mL注入小鼠右侧腋下皮下.肉眼观察并通过核磁共振成像(MRI)测量小鼠成瘤情况,切片光镜观察瘤体组织结构.结果:NOD scid小鼠原位及皮下均能成瘤,MRI及组织病理切片结果均提示种瘤成功.结论:经卵巢原位注射A2780-cp细胞能有效地建立小鼠卵巢癌原位成瘤模型.MRI能无创性检测小鼠卵巢癌成瘤情况.
Aim: To establish A2780-cp ovarian cancer cell lines in situ tumor model on NOD scid mice, in order to provide the reference for the further study of the carrier for ovarian cancer. Methods Cultivation, collection and PBS wash cells and made into cell density of 5 × 10^6/mL single cell suspen-sion in vitro. In situ plant: took 20 μL cell suspension injecting in NOD scid mice ovarian subcutaneous. Subcutaneous planting : 0. 1 mL cell suspension injected in right axillary subcutaneous of the mice. Ob-servation and through MRI measured tumor in mice, and used the HE staining to observe tumors present organization structure slice. Results: Both in situ and subcutaneous, SCID NOD mice can be discovered tumour mass. MRI and histopathology Can be used to confirm this conclusion. Conclusion : Ovarian carci-noma in SCID NOD mice can be effectively established by ovarian in situ injection of A2780-cp cells. MRI can be used to detect the tumor of ovarian cancer in mice.
