针刺对应激性高血压前期大鼠肾脏基因表达的影响 被引量:6

摘要 目的以应激性高血压前期大鼠为模型,运用基因芯片技术和生物信息学分析技术,研究针刺对其肾脏基因表达的影响。方法将9周龄雄性Wistar大鼠,随机分为空白组、模型组、针刺组。模型组和针刺组运用噪声结合足底电击刺激的造摸方式,造模同时,针刺组针刺曲池穴、太冲穴进行干预,模型组和空白组每天只做与针刺组相同的抓取刺激。记录在造模前1天、第3、5、7、9、11天大鼠的血压变化,11 d后取大鼠左肾。运用Gene Chip Rat Gene 2.0 ST芯片对应激性高血压前期模型大鼠肾组织进行基因表达谱的检测,分别找出空白组与模型组之间及模型组与针刺组之间的差异表达基因。运用GO数据库对各组间差异表达基因进行基因功能富集分析。结果造模开始后,空白组血压基本稳定;第3天开始模型组血压开始上升,与正常组比较有统计学意义(P<0.01),并出现行为改变。针刺干预第5天开始,针刺组血压低于模型组(P<0.05)。芯片分析显示:与空白组相比,模型组共筛选出的差异表达基因有46个上调,有48个下调;与模型组相比,针刺治疗后差异表达基因有11个上调,有45个下调;基因功能富集分析显示差异基因与生物调节,刺激反应,多细胞生物体过程,代谢过程,免疫系统进程,受体活性,调节酶,转运蛋白活性等功能有关。结论针刺太冲、曲池穴对于应激性高血压前期大鼠具有一定的降压效应,并可能通过下调Cntn4,Spta1,Mir10,Fbxo15基因来调节血压并对其早期肾脏损害有一定的保护作用。 Objective To use gene chip technology and bioinformatics technologies,study the effects of acupuncture on gene expression in kidney of stress-induced pre-hypertension rat. Methods The 9-week-old male Wistar rats were randomly divided into blank group,model group,and acupuncture group. Model group and acupuncture group using noise combined with foot shock to stimulate touch,at the same time,the acupuncture group was applied to bilateral "Taichong"(LR3) and Quchi(LI11) for 20 min. The model control and blank control group rats were grabbed in the same restrainers for 20 min without any other intervene. Recorded in model 1 day before,3 th 5 th 7 th 9 th 11 th day rats blood pressure changes,after 11 days,draw left kidney materials of rats and use Gene Chip Rat Gene 2. 0 ST chips analysed the differentially expressed genes between the blank group and model group,model group and acupuncture group. Using the differentially expressed genes between groups for enrichment of gene function analysis by GO database. Results Themodel began,the blank group blood pressure remain stable;3rd day of model group blood pressure begins to rise,significant differences compared with the normal group( P 〈0. 01),and behavioural change. 5th day the acupuncture group blood pressure lower than the model group(P 〈0. 05). Microarray analysis showed that compared with the blank group,model group of the differentially expressed genes screened out 46 increases 48 reduced,after acupuncture treatment there are 11 increase and 45 down of differentially expressed genes;Enrichment of gene function analysis revealed differences in biological regulation,response to stimulus,genetic and biological regulation,multicellular organismal process,metabolic process,immune system process,enzyme regulator activity,transporter activity and other function. Conclusion Acupuncture at Taichong and Quchi can lower the SBP of stress-induced pre-hypertension rats and possibly through down the genes Cntn4,Spta1,Mir10 a,Fbxo15 to regulation blood pressure and protect kidney.
