补气助阳汤对变应性鼻炎模型大鼠TNF-α、NF-kB影响随机平行对照研究

Buqi Zhuyang Decoction(补气助阳汤) on Allergic Rhinitis Rats TNF-a, NF-kB Effect of Randomized Controlled Study

[目的]观测补气助阳汤对过敏性鼻炎模型肿瘤坏死因子-α、核因子-κB细胞因子表达影响。[方法]使用随机平行对照方法,将30只SPF级SD大鼠按随机数字表法随机分为三组,空白对照组(A组),模型对照组(B组),补气助阳汤组(C组),10只/组。使用卵蛋白腹腔注射进行致敏,局部鼻黏膜激发的方法复制过敏性鼻炎大鼠模型。除A组外,其余各组大鼠均以卵蛋白溶液给大鼠进行腹腔注射进行致敏,注射剂量为1m L/只,隔日1次,共注射7次。实验第15d,除A组外的其余两组大鼠均以卵蛋白溶液10μL进行滴鼻激发,共11d,每日滴鼻一次。A组大鼠不做任何处理,正常饲养;B组大鼠给予生理盐水从口灌胃,剂量为2m L/只;C组大鼠补气助阳汤中药汤剂经口灌胃,剂量为2m L/只;自激发之日起(实验第15d)开始给药,直至激发结束,共给药11d,1次/d。采用HE染色的方法,观察各组大鼠鼻黏膜病理变化;western-blot法,对各组大鼠鼻黏膜组织中TNF-α、NF-κB的蛋白表达水平进行检测。[结果]与空白对照组比较,其余大鼠鼻黏膜TNF-α、NF-κB含量均显著升高(P<0.01),与模型对照组比较,补气助阳汤组鼻黏膜TNF-α、NF-κB含量有降低(P<0.01)。[结论]补气助阳汤对过敏性鼻炎治疗效果良好,能够抑制患病大鼠鼻黏膜组织内的NF-κB和TNF-α表达水平,发挥了抗过敏作用。

[Objective] Observation Buqi Zhuyang Decoction on allergic rhinitis model of tumor necrosis factor alpha, nuclear factor kappa B cell factor expression in rats. [Methods] Used random parallel control method, 40 SD rats of SPF grade according to the random number table were divided into three groups, blank control group （group A）, model group （B group）, Buqi Zhuyang Decoction group （Group C）, 10 rats/group. The use of egg protein intraperitoneal injection for sensitization, local nasal mucosa excitation method of allergic rhinitis rat model replication. Except for group A and the rest of the rats were with ovalbumin （OVA） solution to rats were intraperitoneal injection were sensitized, injection quantity for the LML/, once every other day, a total of seven injections. Experiment 15d, in addition to the rest of the two groups of rats in the group of rats were egg protein solution 10 L nasal drops, a total of 11d, a daily nasal drops. Rats in group A did not do any treatment, normal feeding. Group a rats without any treatment, normal feeding; rats in group B were given normal saline by gavage dose of 2 mUonly; group C rats Buqi Zhuyang Decoction oral gavage dose of 2 mL/only; since the excitation （the 15d） began administration until the excitation end, a total of 11 days to medicine, 1 times daily. HE staining was used to observe the pathological changes of nasal mueosa of rats in each group. Western-blot method was used to detect the protein expression of TNF- alpha and NF- nuclear factor kappa B in the nasal mucosa of rats in each group. [Results] And compared with the control group, remaining in rat nasal mueosa of TNF-α, NF kB in increased significantly （P〈0.01）, compared with the model group, Buqi Zhuyang Decoction group of nasal mucosa of TNF-a, NF kB in have decreased （P〈0.01）. [Conclusion] Buqi Zhuyang Decoction on allergic rhinitis curative effect good, can inhibit the prevalence rat nasal mucosa tissue within the NF-kB and TNF -α expression level of play anti allergic effects.
