电针镇痛有效群和无效群大鼠下丘脑和PAG阿片肽降解酶蛋白表达的比较研究 被引量:1

摘要 [目的]通过检测针刺镇痛有效群、无效群大鼠下丘脑(hypothalamus,HYPO)和中脑导水管周围灰质(periaqueductal gray,PAG)中阿片肽降解酶中性肽链内切酶(neutral endopeptidase,NEP)与氨基肽酶N(aminopeptidase,APN)蛋白表达的差异,探讨电针镇痛的个体差异。[方法]选用40只SD雄性大鼠,通过检测电针前后甩尾潜伏期(tail flick latency,TFL)变化,筛选出电针有效群与无效群大鼠,有效群9只,无效群20只。两组大鼠适应性喂养一周后,再行一次电针治疗后立即处死,取大鼠下丘脑与PAG,采用免疫印迹法检测NEP和APN的蛋白表达情况。[结果]无效群大鼠下丘脑与PAG中NEP与APN蛋白表达均显著高于有效群大鼠,差异具有统计学意义(P<0.01)。[结论]下丘脑和PAG内阿片肽降解酶可能是介导针刺镇痛无效的主要物质。 [Objective] By detection of the protein expression of neutral endopeptidase(NEP) and aminopeptidase(APN) in hypothalamus(Hypo) and periaqueductal gray(PAG) of the response group and the non-response group of acupuncture analgesia, to investigate the individual differences in electroacupuncture analgesia. [Methods] 40 male Sprague-Dawley rats were used in this study to screen 9 rats in response group and 20 rats in non-response group through the changes in tail flick latency(TFL) before and after electroacupuncture. Two groups of rats were fed a week after adaptive feeding, and sacrificed immediately after the treatment of electroacupuncture only once. The hypothalamus and PAG were removed to detect the protein expressions of APN and NEP, using western blotting. [Results]The protein expression of NEP and APN in hypothalamus and PAG of non-response group rats were significantly higher than those of response group(P〈0.01). [Conclusion]Opioid peptide degradation enzyme in hypothalamus and PAG may be main substances which mediate ineffective electroacupuncture analgesia.
