色氨酸缺乏及外源性吲哚对沙眼衣原体生长的影响 被引量:1

摘要 目的探讨色氨酸缺乏及外源性吲哚对沙眼衣原体E-UW-5/Cx和D-UW-5/Cx型标准株及临床株生长的影响。方法分别用DMEM-10细胞培养液(简称:DMEM-10)、缺乏色氨酸的DMEM-10细胞培养液[简称:DMEM-10(-Trp)]和缺乏色氨酸的DMEM-10细胞培养液中加入10μM外源性吲哚的细胞培养液[简称:DMEM-10(-Trp)+IND]三种液体培养沙眼衣原体至48h,甲醇固定,计数包涵体,观察色氨酸缺乏及外源性吲哚对E型和D型沙眼衣原体标准株及临床株生长的影响。结果 DMEM-10组(-Trp)组衣原体包涵体计数明显低于DMEM-10和DMEM-10(-Trp)+IND组,且差异有统计学意义(P<0.01);但DMEM-10组和DMEM-10(-Trp)+IND组包涵体计数差异无统计学意义(P>0.05);E型标准株、临床株与D型标准株、临床株之间生长差异也无统计学意义(P>0.05)。结论色氨酸缺乏不利于沙眼衣原体的生长,补充外源性吲哚后,沙眼衣原体可恢复其感染活力。 Objective To investigate the effects of tryptophan-starved and exogenous indole on the growth of the Chlamydia trachomatis(Ct) reference strains E-UW-5/Cx, D-UW-5/Cx serotype and the clinical strains. Methods DMEM-10, DMEM without L-tryptophan [ DMEM-10 (-Trp) ] and DMEM-10 (-Trp) added 10txM exogenous indole [ DMEM-10(-Trp) + IND] were utilized incell culture and C. trachomatis infection process, respectively. After incubation for 48 h, the inclusions of Ct were fixed by methanol and counted under optical microscopy using iodine dye staining to observe the effects of tryptophan-starved and exogenous indole on the growth of Ct reference strains E, D serotype and the clinical strains. Results The number of Ct inclusions in DMEM-10(-Tip) group were significantly lower than other groups(P 〈 0.01 ) ; meanwhile, the number of Ct inclusions in DMEM-10 and DMEM-10(-Trp) + IND group represented no significant difference (P 〉0. 05) ; in addition, the Ct reference strains E, D serotype strains and the clinical strains had no significant difference in Ct inclusions numbers (P 〉 0. 05). Condusion Tryptophan-starved status significantly decreased the number of Ct inclusions. And after adding exogenous indole, Ct can restore its ability of infection.
