ADAM33基因F+1多态性、IgE、维生素D与乌鲁木齐地区维吾尔族、汉族哮喘儿童的相关性研究 被引量:3

摘要 目的探讨乌鲁木齐地区维吾尔族、汉族支气管哮喘儿童ADAM33基因F+1位点多态性、Ig E、维生素D(Vit D)水平及其相关性,期望从基因水平为防治儿童支气管哮喘提供理论依据。方法选取2014年2月-2015年6月在新疆医科大学第五附属医院及新疆医科大学第一附属医院门诊及入院支气管哮喘患儿197例(其中汉族111例,维吾尔族86例)作为试验组,同时设立对照组120例(汉族64例,维吾尔族56例),首先采用PCR聚合酶链反应对ADAM 33基因F+1位点进行单核苷酸多态性(SNP)分型,并用ELISA法检测Ig E、Vit D水平,进一步根据民族进行组内分层,进行F+1位点SNP、Ig E、Vit D分析并进行相关性分析。结果 (1)试验组和对照组ADAM33基因F+1位点3种基因型频率分布差异无统计学意义(χ2=0.130,P=0.940),同时等位基因频率差异也无统计学意义(χ2=0.150,P=0.700)。(2)维吾尔族、汉族支气管哮喘儿童中F+1位点分布差异均无统计学意义(χ2=2.955,P=0.228;χ2=1.746,P=0.418)。(3)试验组Ig E-OD水平高于对照组,Vit D-OD水平低于对照组,差异有统计学意义(χ2=13.250,P=0.000;χ2=4.793,P=0.000)。(4)试验组组内维吾尔族支气管哮喘儿童Ig E水平高于汉族儿童,Vit D水平低于汉族支气管哮喘儿童,差异有统计学意义(χ2=2.434,P=0.016;χ2=2.956,P=0.004);对照组组内维吾尔、汉族儿童Ig E、Vit D水平比较差异无统计学意义(χ2=1.072,P=0.286;χ2=0.619,P=0.537)。(5)ADAM 33基因F+1位点与Ig E、Vit D水平均不相关(r=0.050,P=0.377;r=-0.044,P=0.439)。结论 ADAM33基因F+1位点与乌鲁木齐地区维吾尔族、汉族支气管哮喘儿童发病均不相关,ADAM 33基因F+1位点不影响Ig E及Vit D水平。 Objective To investigate the correlation of ADAM33 F+1 gene polymorphism,IgE and Vitamin D levels in asthma children of Uyghur and Han in Urumqi to provide guiding prevention and treat-ment of children with asthma in genetic level.Methods 197 children with asthma (Han 111,Uyghur 86) were selected from February 2014 to June 2015,while a control group of 120 people (Han 64,Uyghur 56),using the polymerase chain reaction and restriction fragment length polymorphism (PCR-RFLP)to analyze ADAM33 F+1 gene single nucleotide polymorphism (SNP),ELISA to measure the IgE and VitD levels,and further analyze the correlation between them based on factor of ethnics.Results (1)The geno-type frequencies of F+1 Locus had no statistical significance between cases and control groups (χ2=0.130, P =0.94),at the allele frequencies had no statistical significance too (χ2=0.15,P=0.70).(2)ADAM33 F+1 in Uyghur and Han asthma children was no statistically significance (χ2 =2.955,P=0.228;χ2=1.746,P =0.418).(3)IgE level was higher while VitD level was lower than the control group,which the differences were statistically significant (χ2=13.250,P=0.000;χ2=4.793,P =0.000).(4)In the case group,Uyghur asthma children IgE level was higher,while VitD level was below than that in Han group, which the differences were statistically significant (χ2=2.434,P=0.016;χ2=2.956,P=0.004);the lev-els of IgE and VitD between Uyghur and Han children in the control group were no significant difference (χ2=1.072,P=0.286;χ2=0.619,P=0.537).(5)ADAM33 F+1 and IgE,VitD had no correlations ob-served (r=0.050,P=0.377;r=-0.044,P=0.439).Conclusion The ploymorphism of ADAM 33 F+1 was not related with asthma of Uyghur and Han children in Urumqi.ADAM33 F+1 did not affect the IgE and VitD levels.
