电针对嗅鞘细胞凋亡的实验研究

摘要 目的探讨电针治疗对嗅鞘细胞在脊髓损伤修复过程中的细胞凋亡影响。方法制备完全离断的脊髓损伤动物模型,造模后分为治疗组和对照组。在造模后1周注射移植细胞,治疗组在细胞移植后给予低频直流电刺激,对照组单纯给予细胞移植。采取H-E染色、免疫荧光染色和Tunel染色等方法观察脊髓损伤的修复程度,明确嗅鞘细胞移植后的存活情况。结果 H-E染色显示治疗组脊髓结构较对照组清晰,神经纤维具有方向性。免疫荧光染色显示治疗组和对照组均有细胞在体内存活,但治疗组的细胞数量多于对照组,二者相比具有统计学意义(P<0.05)。Tunel染色表明治疗组较对照组神经元细胞凋亡的数量少,二者差异具有统计学意义(P<0.05)。结论电针治疗与嗅鞘细胞移植在修复脊髓损伤中能够相互促进和协同,明显减少嗅鞘细胞移植后的凋亡,具有促进嗅鞘细胞在体内存活的作用。 Objective To explore the effect of electro‐acupuncture on apoptosis of olfactory ensheathing cells in repairing spinal cord injury .Methods The animal models of completely amputated spinal cord injury were divided into the treatment groups and the control groups .The olfactory sheath cells were transplan‐ted one week later .The treatment group was treated with low‐frequency galvanic stimulation after cells transplantation ,while the control group was only treated with cell transplantation .The degree of repairing spinal cord injury was observed by H‐E staining ,immunofluorescence staining and Tunel staining ,and the survival rate of olfactory sheath cells after transplantation was calculated . Results H‐E staining results showed that the structures of the spinal cord in the treatment group were more distinct than those in the control group ,and the nerve fibers revealed some directivity .The immunofluorescence staining results showed that some cells in the both groups were alive ,but the number of cells were more in the treatment group than in the control group ( P〈 0 .05) .The Tunel staining results showed that the treatment group had less apoptotic cells the control group ( P 〈 0 .05) .Conclusion Electro‐acupuncture therapy and olfac‐tory sheath cells transplantation can cooperatively promote the repair of spinal cord injury ,can reduce the apoptosis of olfactory ensheathing cells obviously ,and can promote the survival of olfactory sheath cells in vivo .
