摘要
利用ISSR技术对老芒麦(Elymus sibiricus L.)和紫芒披碱草(Elymus purpuraristatus C.P)及其杂种F1代与BC1代的遗传差异性进行研究。结果表明,从90个ISSR引物中筛选出8个适宜引物,共扩增出71个ISSR位点,多态性位点比率为36.6%。供试材料的遗传距离变幅为0.230 8~0.659 1,老芒麦与紫芒披碱草的遗传距离较大(0.712 2),以GD值0.50为基准,分为两类,母本老芒麦、正交F_1代、L-BC_1、Z-BC_1为一类。父本紫芒披碱草、反交F_1代为一类,BC_1代偏向轮回亲本老芒麦遗传。
The genetic differences among Elyrnus sibiricus L., Elymus purpuraristatus C. P, their hybrid F1 and BC1 were investigated by using ISSR technique. The results showed that 71 loci were amplified from 8 primers out of 90 ISSR primers,the polymorphic loci percentage were 36.6%. The genetic distance (GD) of tested plants were ranged from 0.230 8 to 0.659 1 ,and the genetic distance of E.sibiricus and E.purpuraristatus was ulterior(0.712 2), using the GD value of 0.5 as threshold, 6 tested plants could be clustered into 2 groups, group 1 including female parent E. sibiricus and E.sibiricus X E.purpuraristatus F1 ,L-BC1 ,Z-BC1, group 2 including male parent E.purpuraristatus, E.purpuraristatus X E.sibiricus F1 ,BC1 tended to inherit toward recurrent parent E.sibiricus.
