摘要
目的探讨高迁移率蛋白B1(HMGB1)、基质金属蛋白酶-9(MMP-9)m RNA和蛋白在子宫内膜癌组织中表达水平及临床意义。方法应用实时荧光定量RT-PCR及Western印记法检测正常子宫内膜组织、单纯型增生症子宫内膜组织、非典型增生子宫内膜组织和子宫内膜癌Ⅰ期、Ⅱ期及Ⅲ期组织中HMGB1、MMP-9 m RNA和蛋白的表达水平。结果与正常子宫内膜组织比较,子宫内膜癌组织HMGB1、MMP-9 m RNA及蛋白的表达量明显升高,差异有统计学意义(P<0.05);子宫内膜癌组织HMGB1、MMP-9蛋白的表达上调与手术病理分期、淋巴结转移及浸润深度有关(P<0.05),与癌组织的分化程度无关(P>0.05);子宫内膜癌组织中,HMGB1蛋白与MMP-9蛋白的表达呈正相关(r=0.900,P<0.05)。结论 HMGB1、MMP-9在子宫内膜癌的发生、发展及转移的过程中起着重要的作用,其有望成为子宫内膜癌的早期诊断及预后评价的分子标志物。
Objective To investigate theexpression and clinical significance of high mobility group box1 protein (HMGB1) and matrix metalloproteinase-9 (MMP-9) in endometrial carcinoma. Method The expression levels of HMGB1 and MMP-9 were detected by Q-PCR and Western blot in the normal endometrial tissue, endometrial hyperplasia,atypical endometrial hyperplasia, and endometrial cancer (stageⅠ, Ⅱ, Ⅲ ). Results Q-PCR and Western blot results showed that in comparison with normal group ,the expression levels of HMGB1 and MMP-9 in endometrial carcinoma tissues were significantly increased (P〈0.05). The expression levels of HMGB1 and MMP-9 were correlated with FIGO stage, lymph node metastasis and invasion depth (P〈0.05).The expression level of HMGB1 was positively correlated with MMP-9 expression level (r=0.900,P〈0.05). Conclusions HMGB1, MMP-9 may play an important role in the development and metastasis of endometrial carcinoma. It may be able to use as a molecular marker for early diagnosis and prognosis evaluation of the endometrial carcinoma.
