白藜芦醇通过抑制炎症因子表达预防术后腹腔粘连的实验研究 被引量:3

摘要 目的研究白藜芦醇是否能通过抑制炎症反应预防术后腹腔粘连形成。方法通过开腹术后擦拭腹膜诱导粘连形成。SD大鼠32只分为4组:单纯开腹组、阳性对照组、透明质酸钠组和白藜芦醇组。手术后第8天,通过肉眼观察法进行粘连分级评估;通过组织天狼星红染色评估粘连组织间沉积胶原量;通过酶联免疫吸附法检测血清中白介素-6(IL-6)和转化生长因子-β1(TGF-β1)水平;通过免疫印迹试验检测炎症相关分子环氧合酶-2(COX-2)在大鼠腹膜或粘连组织中的表达。结果白藜芦醇显著减轻大鼠模型的腹腔粘连形成,减少纤维蛋白沉积(P<0.05);明显降低血清IL-6和TGF-β1水平(P<0.05);下调COX-2在大鼠腹膜或粘连组织中的表达(P<0.05)。结论白藜芦醇可以有效预防大鼠模型术后腹腔粘连的形成,可能与其通过抑制大鼠损伤腹膜中炎症因子的表达有关;可能是一种有效的新的抗粘连药物,具有潜在应用价值。 Objective To investigate whether the resveratrol can prevent intraperitoneal adhesion formation through inhibiting inflammatory response. Methods The intraperitoneal adhesion was formed by standardized cecal abrasion in Sprague-Dawley rat. Thirty-two rats were randomly divided into 4 groups: simple laparotomy group, positive control group, sodium hyaluronate group and resveratrol group. The extent and tenacity of adhe- sions were evaluated by visual observation at 8 d postoperation. Picrosirius red staining assay was performed to access collagen in tissue sections. Elisa assay was performed to detect the levels of IL-6 and TGF-β1. West- ern blot was used to evaluate the COX-2 protein expression. Results Resveratrol could significantly decrease the formation of intraperitoneal adhesion and collagen deposition (P 〈 0.05). Meanwhile, it could also in- hibit the expressions of serum IL-6 and TGF-β1. The expression of COX-2 could be inhibited by resveratrol. Conclusion Resveratrol can be a new potential agent against intraperitoneal adhesion.
