摘要
目的研究洛铂对人膀胱癌细胞的抑制作用,并揭示其可能的作用机制。方法体外常规培养人膀胱癌细胞株5637。利用MTT细胞增殖法检测洛铂的细胞毒性;采用Annexin V-FITC/PI凋亡试剂盒在流式细胞仪上检测癌细胞的凋亡程度;通过伤口愈合实验观察细胞的迁移;通过Transwell小室测定评估洛铂对5637细胞侵袭和迁移能力的影响;利用蛋白免疫印迹法检测细胞凋亡基因的表达。结果洛铂抑制人膀胱癌细胞并诱导其凋亡,下调膀胱癌5637细胞Survivin和Bcl-2的表达水平,上调其Bax和Caspase-3的表达水平。结论洛铂的细胞毒性诱导人膀胱癌细胞凋亡,有利于洛铂对膀胱癌治疗的深入研究。
Objective To assess the inhibition effect of lobaplatin on human bladder cancer cells,and reveal the possible mechanisms.Methods Bladder cancer cell line 5637 was routinely cultured in vitro.The cytotoxic effect of lobaplatin was determined by MTT cell proliferation assay;Annexin V-FITC/PI KIT was used to detect the degree of apoptosis by flow cytometry;Wound healing assay was conducted to inspect the migration of cells;Transwell chamber assay was completed to assess the effect of lobaplatin on invasion and migration of the bladder cancer cells;western blot was performed to detect the expression of apoptotic gene.Results Lobaplatin can inhibit human bladder cancer cells and induce apoptosis.In addition,lobaplatin can decrease the expression of Survivin and Bcl-2and increase the expression of Bax and Caspase-3in the cell line 5637.Conclusion Lobaplatin cytotoxicity can induce apoptosis of bladder cancer cells,which is helpful to in-depth study on the therapy of bladder cancer using lobaplatin.
