摘要
目的研究绿茶提取物的抗肿瘤活性。方法不同浓度的绿茶提取物作用于MCF-7细胞,通过观察细胞形态,检测细胞周期和凋亡。结果绿茶提取物可以阻滞人乳腺癌细胞株MCF-7生长周期于G1期;荧光染色显示细胞凋亡率增高,电镜观察,出现细胞染色质浓缩、核周边集,线粒体形态正常等细胞凋亡的典型特征;绿茶提取物作用后可以导致MCF-7细胞中Caspase3蛋白活性增强,促进肿瘤细胞的凋亡。结论绿茶提取物对乳腺癌细胞均有一定的抑制作用,进一步筛选其有效成分可以用于肿瘤的预防与治疗。
Objective Green tea extracts have many effects on anti-cancer,but the corresponding mechanism is still exploring.Methods In this experiment,different concentrations of green tea extracts were applied onto MCF-7 cells. Results Typical characteristics of cell apoptosis,such as chromatin condensation,perinuclear clustering and mitochon-dria morphism,were observed in SEM pictures.The flow cytometry detected that green tea extracts can facilitate MCF-7 apoptosis and could retard MCF-7 growth at G1 stage.Fluorescence staining reflects that green tea extracts increases apoptosis rate.Conclusion Therefore,green tea extracts can be used as similar demethylated,low-toxicity and even nontoxic natural plant drugs for antineoplastic.
