银杏叶提取物对急性期溃疡性结肠炎模型小鼠防御素的调节作用

摘要 目的:银杏叶提取物(gingko biloba extract,EGB 761)对急性期溃疡性结肠炎模型小鼠防御素的调节作用。方法:30只小鼠随机分为正常对照组、模型组、EGB761组,建立急性溃疡性结肠炎动物模型;观察小鼠一般情况及结肠病理学变化,应用免疫组织化学染色、Western blot技术检测结肠组织的防御素~1(HBD-1)和防御素~2(HBD-2)的蛋白表达情况。结果:经EGB 761治疗后模型小鼠的一般情况及病理组织学均有明显改变,与模型组比较,EGB 761组结肠组织HBD-1mRNA和蛋白表达明显增加,但HBD-2mRNA和蛋白表达明显下降。结论:可见EGB761对急性期溃疡性结肠炎模型小鼠结肠组织中的防御素具有调节作用。 Objective: To investigate the the effects of Gingko Biloba Extract on the human defensins in the acute experimental colitis. Methods: Thirty mice were randomly grouped as follows: controlled group,model group,EGB 761 group,acute colitis were established,general condition and colon pathological changes were observed,the expressions of HBD- 1and HBD-2mRNAand proteins were detected by real-time Q-PCR,western blot. Results: General condition and colon pathological were changed in the EGB group compared with that of the model group; the proteins and mRNA of expressions of HBD- 1and HBD-2were significantly increased in the EGB group compared with that of the model group by western blot,real-time Q-PCR. Conclusions: The administration of EGB may be adjusted the human defensins in the acute experimental colitis.
