探讨黄芪提取物测定毛蕊异黄酮苷与黄芪甲苷两种成分反相高效液相色谱法研究 被引量:1

摘要 目的:探讨黄芪提取物中两种指标性成分毛蕊异黄酮苷与黄芪甲苷,采用反相高效液相(RP-HPLC)色谱法的测量方法。方法:使用Nobel Kromasil C8,4.6毫米×500毫米×5微米的色谱柱,以每分钟1毫升的流动相洗脱,其流动相按照适当的比例制备,以乙腈兑纯净水制备而成,采用的检测波长分别为240纳米和200纳米。结果:黄芪提取物中的黄芪甲苷毛蕊异黄酮苷这两种成分的峰面积与其进样量含有良好的线性关系,并且它们的相关系数均超过0.9995,仪器的精密度的标准差不超过2%,平均回收率均大于98%。结论:反相高效液相色谱法测定黄芪提取物中的毛蕊异黄酮苷与黄芪甲苷两种指标性成分,速度快,操作简单,回收率高,是定量芪提取物中这两种指标性成分的有效方法。 Objective: To investigate the determination of two components of Calycosin glycoside and Astragaloside IV in Astragali root extract by reversed phase high performance liquid chromatography( RP-HPLC). Methods: The research used nobelkromasil C8column( 4. 6mm×500mm×5U) and the velocity of flow manufactured by the proper proportion was 1ml per minute.The detection wavelength was 240 nm and 200 nm. Results: The sample volume and peakarea of two components of calycosin glycoside and Astragaloside IV in Astragali extract had a perfect linearity. Their correlation coefficients were greater than 0. 9995.Instrument precision of standard deviation was less than 2% and average recovery rate was higher than 98%. Conclusion: The method of RP-HPLC survey and evaluatethe two components of Calycosin glycoside and Astragaloside IV in Astragali root extract is fast,simple operation,high recovery and this method is effective.
