摘要
目的研究电针预处理对大鼠脊髓缺血再灌注损伤后血脊髓屏障的影响。方法 SD大鼠90只,随机分为3组,每组30只:假手术组(S组)、脊髓缺血再灌注损伤组(IR组)、电针预处理组(E组)。IR组和E组大鼠阻断胸主动脉造成缺血再灌注损伤模型,S组只开胸暴露胸主动脉但不阻断,E组在夹闭胸主动脉前行电针刺激预处理。观察记录术后48 h大鼠神经运动功能评分,依文思兰荧光法测定血脊髓屏障通透性,Western blot检测脊髓组织中AQP-4表达情况。结果与S组比较,IR组神经功能评分明显降低,血脊髓屏障通透性明显增加,AQP-4表达明显增多(P<0.05)。与IR组比较,E组神经功能评分增高,血脊髓通透性降低,AQP-4表达减少(P<0.05)。结论电针预处理可以明显抑制脊髓缺血再灌注损伤后AQP-4的表达,降低血脊髓屏障通透性,进而减轻脊髓缺血再灌注损伤。
Objective To study the effects of electroacupuncture preconditioning on the blood spinal cord barrier after spinal cord ischemia reperfusion injury in rats. Methods Ninety SD rats were randomly divided into three groups: sham operation group ( Group S) , spinal cord ischemia reperfusion injury group ( Group IR) , and electroacupuncture pre- conditioning group (Group E). The ischemia reperfusion injury model was established in rats in Group IR and Group E u- sing thoracic aorta clamping. Rats in Group S were only experienced thoracic aorta exposure. Rats in Group E were treated with electroacupuneture pretreatment before injury. Motor function scores were recorded 48h after injury. The permeability of blood spinal cord barrier was determined by Evans blue fluorescence. Western blot was used to detect the expression of AQP -4 in the spinal cord tissues. Results Compared with the Group S, the neurological scores in Group IR were signifi- cantly reduced, with the permeability of blood spinal cord barrier significantly increased, and so was the expression of AQP - 4 ( P 〈 0. 05 ). Compared with Group IR, the neurological scores in Group E were increased, with the permeability of blood spinal cord decreased, and so did the expression of AQP - 4 ( P 〈 0. 05 ). Conclusion Electroacupuncture pre- conditioning can inhibit the expression of AQP -4 after spinal cord ischemia reperfusion injury, which reduce the break- down of blood spinal cord barrier permeability, and thus ameliorate the spinal cord ischemia reperfusion injury.
