鹿茸健骨滴丸对骨质疏松症大鼠肾组织和骨组织CaBP-D_(28k)表达的影响 被引量:1

摘要 目的:本实验采用具有糖皮质激素性骨质疏松症(GIO)的大鼠为实验对象,通过与服用鹿茸健骨滴丸后的大鼠股骨骨密度(BMD)及肾、骨组织CaBP-D_(28k)蛋白表达的比较,探讨糖皮质激素性骨质疏松症的病理机制及鹿茸健骨滴丸的疗效。方法:实验共采用82只Wistar大鼠,并将其随机分为对照组(A组)、模型组(B组)、鹿茸健骨滴丸组(C组)及骨疏康组(D组)。采用后肢肌注地塞米松(2.5 mg·kg^(-1),每周2次)造模,造模同时,C、D组分别按0.88 g·kg^(-1)·d^(-1),2.1 g·kg^(-1)·d^(-1)剂量静脉给药,造模及给药9周后,应用XR-26型双能X线骨密度仪测定BMD,采用实时定量PCR法及Western Blot法测定肾组织和骨组织CaBP-D_(28k)蛋白表达。结果:与正常组比较,模型组大鼠BMD明显降低(P<0.01),肾组织CaBP-D_(28k)蛋白表达明显下降(P<0.01),然而骨组织CaBP-D_(28k)蛋白表达却明显升高(P<0.01);与模空组对比,健骨滴丸组肾组织CaBP-D_(28k)蛋白表达量明显升高(P<0.01),骨疏康组CaBP-D_(28k)蛋白表达量虽有升高趋势,但无统计学意义(P>0.05)。骨组织CaBP-D_(28k)蛋白表达与模空组比较,健骨滴丸组蛋白显著降低(P<0.01),骨疏康组明显降低(P<0.05)。结论:地塞米松是诱发GIO的重要病理机制之一,其对肾钙转运过程产生抑制作用,鹿茸健骨滴丸和骨疏康均通过下调骨钙转运过程的相关蛋白表达,进而发挥抑制骨吸收的作用,达到治疗GIO的作用。 Objective:To observe the effects of Lurong Jiangu Dripping Pills on the bone mineral density (BMD) of femur and the expression of CaBP - D28k in kidney and bone of rats with glucocorticoid - induced osteoporosis (GIO) and research the mechanism of GIO and the therapeutic effect of Lurong Jiangu Dripping Pills. Methods : Eighty - two Wistar rats were randomly divided into normal group ( A), model group ( B), Lurong Jiangu Dripping Pills group (C) and Gushukang group (D). By intramuscular injection of dexamethasone (2.5 mg · kg^-1 ) twice a week to replicate rat models. Different treatment was lasted for 9 weeks simultaneously, such as C group 0. 88 g · kg^-1 · d^- 1, D group 2.1 g · kg^-1· d ^- 1 Detect BMD of femur in vitro with dual - energy X - ray absorptiometry and detect the mRNA and protein expression of CaBP - D28k in kidney by Real - time quantitative RT - PCR and Western Blotting methods. Results:Comparing with the normal group,BMD of the model group decreased evidently (P 〈 0. 01 ), protein expression of CaBP - D28k in kidney decreased evidently (P 〈 0. 01 ), protein expression of CaBP - D28k in bone increased evidently(P 〈0. 01 ). Comparing with the model group, protein expression of CaBP -D28k in kidney of Jiangu Dripping Pills group increased evidently (P 〈 0. 01 ). Although protein expression of CaBP - D28k in kidney of Gushukang group increased, but no statistical significance (P 〉 0. 05 ). Comparing with the model group, Lurong Jiangu Dripping Pills group protein expression of CaBP - D28k decreased evidently (P 〈 0. 01 ). Gushukang group' s protein expression decreased evidently (P 〈 0. 05 ). Conclusion: The depressant effect of dexamethasone for the calcium transportation in kidney was the main pathomechanism of GIO. These two kinds Of traditional Chinese medicine through the reduction of bone calcium transport process related protein expression and play a role in inhibiting bone resorption to achieve the role of the treatment of GIO.
