摘要
目的:观察肉桂提取物肉桂多酚对高脂喂养的C57BL/J6小鼠肝脏脂质沉积的影响并探讨其作用机制。方法:将6周龄雄性C57BL/J6小鼠分为正常对照组(NC组)、高脂组(HF组)和HF+肉桂组(高脂饮食喂养的同时予肉桂多酚200 mg/kg灌胃),喂养8周后处死小鼠取血清测FBG、FINS、ALT、AST、TG、TC水平,取肝脏组织分别用油红O定性及甘油三酯试剂盒定量的方法检测不同实验组肝内甘油三酯的含量,提取肝脏mRNA用q-PCR方法检测各个实验组与脂肪酸从头合成及β有氧相关基因表达水平的改变。结果:与NC组相比,HF组的血清FBG、FINS、AST、TG、TC均升高,肝脏脂质沉积明显,SREBP-1、FAS、SCD1等基因mRNA表达水平升高,PPARɑ、CPT1ɑ、MCAD等基因mRNA表达水平下降;而肉桂组除TC外其余血清学指标较HF组均明显下降(P<0.05),肝脏内脂质沉积较HF组也明显改善,定量后有统计学差异(P<0.05),SREBP-1、FAS、SCD1等基因mRNA的表达较HF组下降,而PPARɑ、CPT1ɑ、MCAD等基因mRNA的表达升高。结论:肉桂多酚可有效改善高脂喂养的C57BL/J6小鼠肝脏脂质沉积,该作用可能是通过抑制脂肪酸从头合成途径,同时促进脂肪酸的β有氧过程而实现的。
Objective : To explore the function of cinnamon polyphenol on liver lipid metabolism and its molecular mechanism in C57BL/6 mice which were fed by high.fat diet. Methods :Six -week -old male C57BL/6 mice were divided into normal control group ( NC group) ,high fat group ( HF group) and HF + cinnamon group ( high fat diet + cinnamon polyphenol 200 mg/kg). The serum levels of FBG, FINS, ALT, AST, TG and TC were determined after 8 weeks. The liver tissues were stained with oil red O and quantified by triglyceride kit to detect the contents of triglyeerides in liver. The mRNA levels of genes related to de novo synthesis and β - oxidation of fatty acid were detected by q - PCR. Results : Compared with the NC group, the serum levels of FBG, FINS, AST,TG,TC ,the lipid deposition of liver and the genes expression of SREBP- 1 ,FAS and SCD1 in HF group were significantly higher than those of NC group (P 〈 0.05 ) while the genes expressions of PPARu, CPT1α and MCAD were decreased (P 〈 0. 05 ). Compared with the HF group, the serum levels of FBG, FINS, AST, TG, the lipid deposition of liver and the genes expression of SREBP - 1, FAS and SCD1 in HF + cinnamon group were significantly decreased than those of HF group ( P 〈 0.05 ) while the genes expressions of PPARu, CPT1 u and MCAD were increased ( P 〈 0. 05 ). Conclusion : Cinnamon polyphenols can effectively improve the lipid deposition in liver of C57BL/6 mice fed with high fat diet, which may be achieved by inhibiting the de novo synthesis pathway and promoting the B oxidation of fatty acids.
