摘要
目的观察裸角质膜同源蛋白(NKD1)、β-连环素(β-catenin)和细胞周期蛋白D1(CyclinD1)在结直肠癌组织中的表达.探讨其在结直肠癌变过程中的意义。方法收集2014年1月~2016年1月在鄂东医疗集团黄石市中心医院(我院)行结直肠癌根治术切除的癌症组织96例及27例肠管正常黏膜,并收集同期在我院治疗的38例结直肠腺瘤切除标本,采用sP免疫组化法对所有标本中NKDl、β—catenin和CyclinD1的表达情况进行检测,分析NKDl阳性率、CyclinD1阳性率与β—catenin异位表达率三者之间的相关性。结果结直肠癌组织中NKDl的阳性率(17.71%)明显低于结直肠正常黏膜(92.59%)及腺瘤组织(78.95%),差异有统计学意义(均P〈0.05);结直肠癌组织中的β—catenin异位表达率(78.13%)和CyclinD1阳性率(84.38%)均明显高于结直肠正常黏膜、腺瘤组织中的表达率(42.11%、34.21%),差异有统计学意义(均P〈0.05);NKDl、β—catenin和CyclinD1在结直肠腺癌组织中的表达均与癌分化程度、Duke分期、淋巴结转移有关(P〈0.05)。结论NKDl、β—catenin和CyclinD1可能参与了结直肠癌的发生、发展、侵袭和转移,联合检测可判断结直肠癌的生物学行为并作为临床治疗的参考依据。
Objective To observe the expression of naked cutin membrane homologous protein (NKD1), β serial element (β- catenin) and cell cycle protein D1 (Cyclin D1) in colorectal cancer tissues and explore its significance in the pro- cess of colorectal cancer. Methods 96 cases of colorectal cancer and intestinal resection of the cancer tissue margin of normal mucosa in 27 cases, 38 cases of colorectal adenoma resection specimens in Huangshi Central Hospital of Edong Healthcare Group from January 2014 to January 2016 were collected. SP immunohistochemical method was used to de- tect the expression of NKD1, beta -catenin and Cyclin D1 in all specimens. The correlation between NKD1 positive rate, Cyclin D1 positive rate and ectopic expression rate of -catenin was analyzed. Results The positive rate of NKD1 in colorectal cancer tissues (17.71%) was significantly lower than that in normal colorectal mucosa and adenoma respec- tively (92.59%, 78.95%), the differences were statistically significant (P 〈 0.05); β-catenin protein expression in colorectal cancer tissues (78.13%) and the rate of Cyclin D1 positive rate (84.38%) was significantly higher than that of normal colorectal mucosa, adenoma tissues respectively (42.11%, 34.21%), the differences were statistically significant (P 〈 0.05); the expression of NKD1, 13 -catenin and Cyclin D1 in colorectal cancer tissues was related to fhe differentiation degree, Duke stage and lymph node metastasis (P 〈 0.05). Conclusion NKD1, β-catenin and Cyclin D1 may be in- volved in the occurrence, development, invasion and metastasis of colorectal cancer, and the combined detection of the biological behavior of colorectal cancer can be used as a reference for clinical treatment.
