摘要
目的探讨林下参花挥发油化学组成及其对多种肿瘤细胞增殖活性的影响.方法通过水蒸气蒸馏法制备林下参花挥发油,应用GC-MS方法鉴定其化学成分,利用MTT法研究其对小鼠成纤维细胞NIH-3T3毒性及对人肺腺癌细胞A549、人宫颈癌细胞He La、人胃癌细胞SGC7901、小鼠黑色素瘤细胞B16和小鼠肾上腺皮质瘤细胞Y1增殖的影响.结果从林下参花挥发油鉴定出85种成分,包括脂肪族、萜类、芳香族等化合物,含量最高为棕榈酸、亚油酸、十三酸等;其在400μg/m L以下对于NIH-3T3细胞生长无毒性作用,且能够抑制Y1,He La,SGC-7901,B16细胞系增殖,对A549细胞系增殖无影响.结论林下参花挥发油化学成分含量较高的是脂肪族和萜类化合物,并且发现其对多种肿瘤细胞系具有抑制增殖活性.
Objective To investigate the chemical composition and anti-proliferations of essential oil from flowers of ginseng under forest( GUF). Method Essential oil of GUF flowers( GUFFEO) was extracted by hydrodistillation and analyzed by GC-MS respectively.The cytotoxicities of GUFFEO on mouse fibroblasts( NIH-3T3) and the effects of GUFFEO on the proliferation of human lung adenocarcinoma( A549),human cervical cancer( He La),human gastric cancer( SGC7901),mouse melanoma( B16) and adrenal cortical( Y1) cell lines were assayed by MTT. Results 85 components of GUFFEO were identified by GC-MS. It contained a complex mixture consisting of aliphatic,terpenoid and aromatic compounds. The major constituents were palmitic acid,linoleic acid and tridecanoic acid. There was no cytotoxicity of GUFFEO on the 3T3 cell lines with the concentration under 400 μg / m L. GUFFEO exhibited significant inhibition of Y1,He La,SGC-7901 and B16 cancer cells growth,and no effect on A549 cell proliferation.Conclusion The chemical composition of essential oil from flowers of ginseng under forest mainly contained aliphatic and terpenoid compounds,and it was found that it can inhibit the proliferations of many cancer cell lines.
