甲型流感病毒感染小鼠肺组织中环状RNA的表达变化及其靶基因功能分析 被引量:3

摘要 目的明确参与甲型流感病毒(IAV)感染小鼠肺组织中环状RNA(circRNA)的表达变化,并分析其靶基因生物学功能。方法采用高通量测序的方法,测定并鉴定了IAV感染组和正常对照组小鼠肺组织中circRNA的表达差异及其靶基因。利用GeneOntology(GO)和KyotoEncyclopediaofGenesandGenomes(KEGG)软件分析差异表达的circRNA靶基因生物学功能。结果共获得12个显著差异表达的circRNAs(差异倍数〉2),其中circRNA5828表达量显著下调,circRNAl3602和circRNAl3853表达量显著上调。生物信息学软件分析表明,12个circRNA靶基因在生物过程、细胞组成和分子功能三方面均发挥作用,并影响抗原加工提呈、T011样受体、维甲酸诱导基因-I样受体和心肌功能等相关信号通路。结论circRNA可能通过宿主免疫调控参与IAV的感染过程。 Objective To investigate differentially expressed circRNAs of lung tissue in mice infected with influenza A virus (IAV) and to analyze biological function of their candidate target genes. Methods The differential expression of circRNAs and their target genes in lung tissues between IAV infected mice and normal mice were clarified by high-flux sequencing. The biological functions of differentially expressed circRNA target genes were analyzed by Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG). Results Twelve differentially expressed circRNAs were identified (〉2 times differences). Among them, the expression level of circRNA5828 was obviously down regulated, while the expression levels of circRNA13602 and circRNA13853 were obviously up regulated. The results of bioinformatics analysis showed that a total of 12 target genes played roles in biological process, cellular component and molecular function, and affected signal pathways related to antigen processing and presentation, Toll-like receptor, retinoic add-induced gene- I like receptor, cardiomyopathy and so on. Conclusions CircRNA may participate in host immunoregulation during IAV infection.
