化痰活血方对哮喘小鼠肺泡灌洗液中IL-5、IL-10、TGF-β1和MUC5AC的影响 被引量:7

摘要 目的研究化痰活血方(Hua-Tan-Huo-Xue-Fang,HTHXF)对卵清蛋白(OVA)引起的哮喘小鼠肺泡灌洗液(Bronchoalveolar lavage fluids,BALF)中白细胞数目及IL-5、IL-10、TGF-β1、MUC5AC表达的影响,并探究其治疗哮喘的可能作用机制。方法腹腔注射含有OVA与氢氧化铝的致敏液致敏,雾化吸入OVA激发哮喘模型。于实验第9天灌胃给药,雾化激发期间灌胃时间控制在雾化激发前1h。实验第28天处死小鼠,并取BALF测定其中的白细胞数目及IL-5、IL-10、TGF-β1和MUC5AC水平。结果化痰活血方能明显降低BALF中淋巴细胞、中性粒细胞、嗜酸性粒细胞、单核细胞和白细胞总数,降低IL-5和MUC5AC的水平,升高IL-10和TGF-β1水平。结论化痰活血方治疗哮喘的作用机制可能与化痰活血方减少肺泡中白细胞的聚集,升高IL-10和TGF-β1,抑制IL-5和MUC5AC的产生,抑制气道黏液高分泌和炎症有关。 Objective To investigate the effects of Hua- Tan- Huo- Xue- Fang(HTHXF) on the amount of white blood cells and the levels of IL- 5, IL - 10, TGF - β1 and MUC5AC in bronchoalveolar lavage fluids (BALF) of OVA -induced asthmatic mice, and then to explore the possible mechanism in asthma therapy. Methods Asthmatic model of mice was sensitized by intrap- eritoneal injection suspension of OVA and A1 (OH) 3, and then aerosol inhalation of OVA was given to stimulate asthma. At the 9th day, drugs were intervened by intragastrieal administration, and insure giving administration of drugs by garage 1 hour before the sensitization of OVA during the period of atomization. At 28th day, sacrificed all the mice and gained BALF in order to meas- ure the amount of white blood cells and the level of IL -5, IL - 10,TGF - β1 and MUC5AC. Results HTHXF can down -regulate the number of lymphocytes, neutrophils, eosinophils, monocytes and white blood cellsin the BALF, apparently reduce the level of IL-5 and MUC5AC in the BALF, improve the level of TGF-β and IL- 10 in the BALF. Conclusion The functional mecha- nism of HTHXF in treating asthma may be HTHXF reduce the recruitment of white blood cells, rise IL - 10 and TGF - β1 ,inhibit the production of IL -5 and MUCSAC, inhibit the hypersecretion of airway mucus and inflammation.
