丹溪痛风方对胰腺癌BxPC-3细胞Hedgehog信号通路关键蛋白的影响 被引量:4

The Effects of Danxi Gout Prescription on Hedgehog Signaling Pathway Proteins of BxPC-3 Cell Line in the Pancreatic Cancer

摘要 目的探讨丹溪痛风方(DGP)对胰腺癌BxPC-3细胞Hedgehog信号通路关键蛋白的影响。方法胰腺癌BxPC-3细胞随机分成模型组,5-氟尿嘧啶(5-FU)组(50μg/ml),DGP高、中、低剂量组(40,20,10μg/ml)。采用台盼蓝染色、MTT法、细胞划痕法分别检测胰腺癌BxPC-3细胞生长曲线、增殖抑制率、运动能力;采用免疫组织化学、RT-PCR法检测胰腺癌BxPC-3细胞Hedgehog信号通路关键蛋白Shh、Smo、Ptch1、Gli1水平。结果与模型组比较,5-FU组,DGP高、中剂量组胰腺癌BxPC-3细胞OD值明显降低(P<0.05);运动能力明显降低(P<0.05);Hedgehog信号通路关键蛋白Shh、Smo、Ptch1、Gli1水平明显降低(P<0.05);Hedgehog信号通路关键蛋白Shh、Smo、Ptch1、Gli1 mRNA水平明显降低(P<0.05)。结论 DGP对胰腺癌BxPC-3细胞Hedgehog信号通路关键蛋白Shh、Smo、Ptch1、Gli1具有抑制作用,可见DGP在胰腺癌治疗中扮演重要角色。 Objective To study the effects of Danxi Gout Prescription(DGP) on Hedgehog signaling pathway proteins of BxPC - 3 cell line in the pancreatic cancer. Methods BxPC -3 cell line of pancreatic cancer were randomly divided into model group,5 - Fluorouracil (5 - FU) group (50μg/ml), DGP high - dose group (40 μg/ml), DGP dose group ( 20 μg/ml), DGP low - dose group ( 10μg/ml). The trypan blue staining and MTT was used to determine growth curve of BxPC -3 cell line and OD values of pancreatic cancer. The cell scratch was used to determine the exercise capacity of BxPC - 3 cell line of pancreatic cancer. The Immunohistochemistry and Real -time -PCR was applied for detecting Hedgehog signaling pathway proteins Shh, Smo, Ptchl, Glil levels in BxPC -3 cell line of pancreatic cancer. Results Compared with the model group,5 -FU group, DGP high -dose group and DGP dose group OD values of BxPC - 3 cell line of pancreatic cancer significantly decreased ( P 〈 0.05), exercise capacity significantly decreased (P 〈0.05 ). Compared with the model group,5 -FU group, DGP high- dose group and DGP dose group BxPC -3 cell line of pancreatic cancer Hedgehog signaling pathway proteins Shh, Smo, Ptchl, Glil protein and mRNA level were significantly decreased(P 〈 0.05 ). Conclusion DGP plays a certain role in the treatment of BxPC- 3 cell line of pancreatic cancer through inhibiting Hedgehog signaling pathway proteins Shh, Smo, Ptchl, Glil.
