TSP-1活化TGF-β_1在糖尿病肾病瘀血阻络证大鼠的表达及化瘀通络中药的干预作用

The activation of TSP-1,TGF-β_1 expression and intervention effect of Chinese Herbs for Stasis Removing and Collaterals on diabetic nephropathy of blood stasis syndrome in rats

目的探讨TSP-1活化TGF-β_1在糖尿病肾病瘀血阻络证大鼠的表达及化瘀通络中药的干预作用。方法 50只雄性SD大鼠,随机选取10只为正常对照组(C组),余40只采用高糖高脂饲料饲养联合小剂量STZ腹腔注射造模。成模大鼠随机分为模型组(M组)、中药组(Z组)、厄贝沙坦组(I组),相应药物剂量灌胃。16周末测空腹血糖(FBG)、糖化血红蛋白(HbA1c)、留取尿液测24h尿微量白蛋白(U-mAlb)、计算右肾重/体重(KI)、PCR及western-blotting检测肾皮质TSP-1、TGF-β_1表达。结果与C组比较,M组FBG、HbA1c、KI、U-mAlb、TSP-1、TGF-β_1 mRNA及蛋白表达水平均明显升高(P<0.05);与M组比较,I、Z两组KI、U-mAlb、TSP-1、TGF-β_1mRNA及蛋白表达水平明显下降(P<0.05)。结论 TSP-1、TGF-β_1可能作为靶点之一参与了化瘀通络中药对糖尿病肾病大鼠的肾保护作用。

Objective To investigate the activation of TSP - 1, TGF - β1 expression and intervention effect of Chinese Herbs for Stasis Removing and Collaterals on diabetic nephropathy of blood stasis syndrome in rats. Methods Fifty male SD rats were ran- domly selected as normal control group （ group C）, and the remaining 40 rats were fed with high fat diet and low dose STZ intrap- eritoneal injection. The model rats were randomly divided into model group （group M）, Chinese medicine group （group Z） and Maher Irbesartan group （group I）. 16 weeks measured fasting blood glucose （FBG）, glycosylated hemoglobin （HbA1 c）, leavethe urine test 24h urinary microalbumin （U -mAlb）, calculation fight kidney weight/body weight （KI）, PCR and Western- blotting were used to detect expression of TSP- 1, TGF- β1 in renal cortical. Results Compared with C group, FBG, HbAlc, KI, U - mAlb, TSP - 1, TGF - β1 mRNA and protein expression levels in M group were significandy increased （ P 〈 0.05 ） ; Compared with M group, I and Z group of KI, U -mAlb, TSP - 1, TGF - β1 mRNA and protein expression levels were signifi- cantly decreased （ P 〈 0.05 ）. Conclusion TSP - 1 and TGF - β1 may be used one of the targets in the protective effect of tradi- tional Chinese medicine on diabetic nephropathy rats.
