高效液相色谱法同时测定改善睡眠颗粒中天麻素、红景天苷、栀子苷和斯皮诺素含量的应用 被引量:1

摘要 目的 建立高效液相色谱同时测定“改善睡眠颗粒”中天麻素、红景天苷、栀子苷和斯皮诺素含量的方法 。方法检测对象为以上提到的“改善睡眠颗粒”中的四个主要成分。采用的色谱柱为Thermo Fisher Syncronis C18(250 mm×4.6 mm,5μm);流动相为乙腈-水,梯度洗脱,流速为1.0 ml/min;柱温为30℃,进样量为10μl。检测波长分别设定为220、238、275、335 nm。结果 天麻素、红景天苷、栀子苷和斯皮诺素分别在0.8032-4.0160、0.5392-2.6960、2.6916-13.4580、0.2112-1.0560μg范围内线性关系良好;精密度、稳定性、重复性试验的均RSD〈2.00%;加样回收率分别为98.17%、99.37%、98.84%、99.27%;RSD分别为0.67%、0.48%、0.49%、0.93%。结论 高效液相色谱法可同时测定“改善睡眠颗粒”中天麻素、红景天苷、栀子苷和斯皮诺素的含量,且该方法简便、准确,可用于该制剂的质量控制。 Objective To establish an HPLC method for simultaneous determination of four constituents (Gastordine, Salidroside,Geniposide Spinosin) in "Gaishanshuimian Granules".Methods Aforementioned four major constituents in "Gais- hanshuimian Granules" as the detecting subject.Column was Thermo Fisher Syncronis C18 (250 mm×4.6 mm,5 μm).Mobile phase was aeetonitrile-water, gradient elute,flow rate was 1.0 ml/min.Column temperature was maintained at 30℃, sample size was 10 pJ.Detection wavelengths were set at 220,238,275 and 335 nm.Results Linear range of Gastordine, Salidroside,Geniposide,and Spinosin respectively were 0.8032-4.0160,0.5392-2.6960,2.6916-13.4580,0.2112-1.0560 gtg. The RSD of precision,stability and repeatability test were all less than 2.00%.The average recoveries respectively were 98.17%,99.37%,98.84% and 99.27% with RSD of 0.67%,0.48%,0.49% and 0.93%,respectively.Conclusion This developed HPLC method is reliable and convenient for simultaneous determination of Gastordine,Salidroside,Geniposide, and Spinosin,which could be applied to determine contents of four constituents and quality control of "Gaishanshuimian Granules".
