艾司洛尔对炎性大鼠脊髓COX-2mRNA表达的影响 被引量:1

摘要 目的 探讨鞘内注射艾司洛尔对甲醛炎性大鼠脊髓背角COX-2 mRNA表达的影响。方法 鞘内置管后,36只SD大鼠采用随机数学表法分为3组(n=12):生理盐水组(N组),300μg/kg(S1组),600μg/kg(S2组)。鞘内置管5 d后,注射以上各组药物,然后在大鼠后肢皮下注射甲醛50μl,以Von-Frey细丝法和Hargreaves法测定机械缩足反射阈值(MWT)和热刺激缩足反射潜伏期(TWL),观察大鼠的疼痛行为学变化。观察1.5 h后,采用RTPCR方法观察脊髓背角L4-5 COX-2 mRNA表达。结果 与N组比较,S1、S2组的Ⅱ相阶段的疼痛行为均有所减少,且S1、S2组L4-5节段脊髓表达的COX-2 mRNA的量均比N组少(P〈0.01)。结论 鞘内注射艾司洛尔可减少甲醛大鼠脊髓背角COX-2 mRNA表达。 Objective To investigate the effect of intrathecal injection of Esmolol on the expression of COX-2 mRNA in the spinal dorsal horn of formaldehyde-induced inflammatory rats.Methods After the intrathecal catheterization,36 SD rats were randomly divided into 3 groups (n=12):saline group (group N),300μg/kg group (group S1),and 600 μg/kg group (group S2).5 days after the intrathecal catheterization,the drug of each group was given to the rats respectively and then 50 μl formaldehyde was injected subcutaneously in the hind limbs of rats.Von-Frey method and Hargreaves method were adopted to measure the mechanical withdrawal threshold (MWT) and thermal withdrawal latency (TWL) so as to observe the pain behavioral changes in rats.After 1.5 hours observation,RT-PCR was given to observe COX-2 mRNA expression in L4-5 spinal dorsal horu.Results Compared with group N,phase II pain behaviors in group S1 and group S2 were reduced and the expression of COX-2 mRNA in L4-5 spinal dorsal horn in group S1 and group S2 was less than that in group N (P〈0.01).Conclusion Intrathecal injection of Esmolol reduces COX-2 mRNA expression in the spinal dorsal horn of formalin-induced rats.
