摘要
目的:测定赶黄草不同体积分数乙醇提取物收率及其中没食子酸含量,为筛选更优的赶黄草提取方法提供参考。方法:1用不同体积分数乙醇采用热回流提取法提取赶黄草,得到赶黄草提取物;2采用Agilent C18色谱柱,甲醇-0.15%磷酸溶液(10∶90)为流动相,检测波长270 nm,柱温35℃,流速1.0 m L·min^(-1)测定赶黄草提取物中没食子酸含量。结果:35%乙醇、55%乙醇、75%乙醇、95%乙醇提取得到的赶黄草浸膏收率分别为16.3%、16.6%、16.8%、9.6%,所得提取物中没食子酸含量分别为10.81μg·mg^(-1)、11.87μg·mg^(-1)、13.61μg·mg^(-1)、15.50μg·mg^(-1)。结论:75%乙醇提取赶黄草收率及提取物中没食子酸含量总量最高,95%乙醇赶黄草提取物中没食子酸浓度最高。
Objective: The content of gallic acid and the recovery rate of Phethorum Chinese Pursh from different concentrations of ethanol extracts were determined in order to provide a better way to recover the Phethorum Chinese Pursh from extraction. Methods: ①We use different volume fractions of ethanol were used in hot reflux extraction to extract the Phethorum Chinese Pursh extraction.②Gallic acid was analyzed by Agilent C18 column at 270nm. The mobile phase consisted of acetonitrile 0.1% phosphoric acid (10:90), running at the velocity at 1.0 mL·min^-1, and the column temperature was 35%.Results: At the concentrations of ethanol of 35%, 55%, 75%, 95% ,the recovery rates of Phethorum Chinese Pursh were 16.3%,. 16.6%, 16.8%, 9.6%, and the contents of gallic acid were 10.81μg-mg^-1, 11.87 μg·mg^-1, 13.61 μg·mg^-1, 15.50μg ·mg^-1, respectively. Conclusion: The recovery rate was the highest using 75% ethanol, whereas 95% ethanol gave the highest concentration of gallic acid.
