miR-100抑制胃癌细胞裸鼠移植瘤的生长及机制的研究 被引量:1

摘要 目的探究miR-100抑制胃癌SGC-7901细胞裸鼠移植瘤的生长及机制。方法将实验裸鼠随机分成miR-100组和阴性对照组,将用GFP-miR-100或GFP-miR-negative control转染的胃癌SGC7901细胞通过裸鼠腋窝注射分别转染到两组裸鼠中去,建立人胃癌SGC7901胃癌miR-100模型和阴性对照组模型,通过GFP图像观察体内miR-100的表达情况,HE染色观察肺转移及基质浸润情况,免疫组织化学和western blot检测miR-100靶基因蛋白表达情况。结果 miR-100在miR-100组裸鼠中稳定地高表达;在阴性对照组的8只裸鼠中,有6只裸鼠的肿瘤细胞浸润肿瘤周围的基质,miR-100组的9只裸鼠中,只有2只出现了基质浸润,其余7只裸鼠中,肿瘤结节被周围的纤维组织完好地包绕。western blot和免疫组织化学显示在裸鼠异种移植肿瘤中,miR-100显著抑制了ZBTB7A蛋白的表达。结论 miR-100通过抑制ZBTB7A靶点基因抑制胃癌细胞裸鼠移植瘤的转移和生长。 Objective To examine the mechanism of miR-100 in inhibiting the growth of human gastric cancinoma SGC-7901 cell in nude mice. Method The nude mice as subjects of the study were first randomly divided into 2 groups: mir-100 group( experimental group) and negative control group. In order to establish the model,SGC7901 cells transfected with GFPmiR-100 or GFP-miR-negative were injected into the axillary of nude mice of the two groups respectively. The expression of in vivo mir-100 by GFP image, lung metastases and stromal infiltration were observed by HE staining.Immunohistochemistry and Western blot were used to detect the expression of target gene of miR-100. Findings miR-100 was overexpressed stably in the experimental group. Six of the eight injected mice in the control group had cancer cells invading the stroma surrounding the tumors. In stark contrast,stromal invasion was only found in two of the nine injected mice in the experimental group. The tumor nodules of the remaining 7 mice were well-encapsulated by intact fibrotic capsules. Western blot and immunohistochemistry showed that miR-100 effectively inhibited the expression of ZBTB7 A protein in xenograft tumor of nude mice. Conclusion miR-100 inhibits the metastasis and growth of human gastric cancer in nude mice by inhibiting the ZBTB7 A target gene.
