血根碱对舌癌体外增殖、凋亡和迁移作用的影响 被引量:2

Effect of sanguinarine on proliferation,apoptosis and migration of tongue squamous cell carcinoma

摘要 目的:研究博落回提取物中血根碱(Sanguinarine,SAN)对舌癌体外增殖、凋亡和迁移作用的影响及其机制。方法:采用不同浓度的血根碱处理Tscca和Tca8113细胞24、48、72 h,MTT法检测血根碱对两种舌癌细胞增殖抑制的影响;用药48 h后,流式细胞仪Annexin V-FITC/PI染色法检测不同浓度血根碱对细胞凋亡的影响;Western blotting法检测不同浓度血根碱对细胞中NF-κB表达的影响;细胞划痕实验检测不同浓度血根碱对舌癌细胞迁移能力的影响。结果:经血根碱处理后的舌癌细胞系Tscca和Tca8113有明显的抑制增殖作用,并呈现一定的浓度和时间依赖关系。血根碱作用于舌鳞癌Tca8113细胞24 h、48 h、72h的半数抑制浓度(IC50)分别为2.53μmol/L、2.38μmol/L、1.96μmol/L。舌鳞癌Tscca细胞24 h、48 h、72h的半数抑制浓度(IC_(50))分别为1.48μmol/L、1.17μmol/L、0.89μmol/L。血根碱可以抑制NF-κB的表达,且有一定的剂量相关性。一定浓度的血根碱还能抑制Tca8113细胞株的迁移。结论:血根碱能显著抑制舌癌细胞Tscca和Tca8113的增殖并且诱导其凋亡,其机制可能与对NF-κB表达的影响有关,同时血根碱可影响Tca8113细胞株的迁移。 Objective: To explore the effects and mechanism of sanguinarine of macleaya cordata extract on proliferation,apoptosis and migration oftongue squamous cell carcinoma. Methods: The inhibitory effects of sanguinarine on the proliferation of tongue cancer cells were determined by MTT assay after Tca8113 and Tscca cells were treated with different concentrations of sanguinarine for 24,48 and 72 hours. The effects of sanguinarine on the apoptosis of tongue cancer cells were detected by flow cytometry of Annexin V-FITC / PI staining method. The expression level of Tongue cancer NF-κB was detected by Western blotting after sanguinarine treatment. The effects of sanguinarine on the migration of tongue cancer cells were detected by cell scratch test. Results: The proliferation of Tca8113 and Tscca cells were significantly inhibited in a time-and concentration-dependent manner after treated by sanguinarine. The half inhibitory concentration( IC50) of sanguinarine on tongue squamous cell Tca8113 are 2. 53μmol / L,2. 38μmol / L and 1. 96μmol / L when the time is 24 h,48 h and 72 h. The half inhibitory concentration( IC50) of sanguinarine on tongue squamous cell Tscca are 1. 48μmol/L、1. 17μmol/L、0. 89μmol/L when the time is 24 h,48 h and72 h. Sanguinarine could inhibit the expression of NF-κB proteins in a concentration-dependent manner. A certain concentration of sanguinarine could inhibit the migration of Tca8113 cells. Conclusion: Sanguinarine can significantly inhibit the proliferation of Tca8113 and Tscca cells and induce apoptosis. The mechanisms might be related with the effect of the expression of NF-κB. It can also inhibit the migration of Tca8113 cells.
