摘要
目的:以Hep G2.2.15细胞系为体外实验模型,研究波棱素脂质体体外抑制乙肝病毒的生物学活性。方法:MTT法检测药物对细胞的毒性,将不同安全浓度的波棱素脂质体及阳性对照药物拉米夫定(3TC)作用于此细胞系,同时实验设不加药物的细胞对照,给药3,6d后收集细胞培养上清液。采用ELISA法测定各组细胞培养上清液中HBs Ag,HBsAg水平,实时定量PCR检测HBV-DNA的含量。结果:波棱素脂质体对HepG2.2.15细胞的TC50为1443.68μg/ml。波棱素脂质体对HepG2.2.15分泌的HBs Ag和HBeAg的IC50分别为131.08μg/ml,109.50μg/ml,治疗指数为11.01和13.18。同时,浓度范围在25~200μg/ml时明显抑制HBV-DNA表达,最高抑制率达到55.04%。结论:波棱素脂质体具有一定的体外抑制乙肝病毒复制及抗原表达的作用。
Objective: In this study,we aimed to investigate the against hepatitis B virus effects of herpetin liposomes in vitro. Method: Hep G2.2. 15 cells were used as experimental model in vitro,the cell toxicity was observed by MTT assay,the different safe concentrations of herpetin liposomes and positive control drug Lamivudine( 3TC) were added into the cells. Experimental set cells control without drug at the same time. Collecting cell culture supernatants after 3days and 6days,HBs Ag and HBe Ag content were detected by ELISA. The content of HBVDNA using fluorescent probe quantitative PCR to detect. Results: TC50 of herpetin liposomes in Hep G2. 2. 15 cells culture was 1443. 68μg /ml. The results indicated drug at 200μg / ml suppressed significantly HBs Ag and HBe Ag expressions on the sixth day. IC50 of herpetin liposomes in Hep G2. 2. 15 cells HBs Ag and HBe Ag were 131. 08μg / ml,109. 50μg / ml,with TI being 11. 01 and 13. 18. Furthermore,herpetin liposomes at 50-200μg / ml also showed an inhibitory effect on HBV DNA replication with maximum inhibitory rate up to 55. 04%. Conclusion: Herpetin liposomes exhibited a significantly effect against HBV replication and antigen expression.
