摘要
建立了反相高效液相色谱(RP-HPLC)测定太白泡沙参中β-谷甾醇含量的方法。在色谱柱为Kromasil C_(18)柱(5μm,4.6mm×250mm)、紫外检测(波长210nm)、色谱柱温度35℃,以甲醇为流动相且流速为0.7mL/min的优化条件下,β-谷甾醇在4×10^(-7)~2×10^(-5)g范围内线性关系良好,r=0.999 9,平均回收率为99.70%,测定太白泡沙参中β-谷甾醇平均含量为0.134%。此分析方法简便准确,可作为该产品的质量控制方法。
A method for the determination of β-sitosterol in A. potaninii Korsh from Shaanxi Taibai Mountainous area by reverse phase-high performance liquid chromatography(RP-HPLC) has been devel- oped. The Kromasil C18 column (4. 6 mm× 250 mm,5 μm) was used for the separation with a mobile phase of pure methanol. The detected wavelength was 210 nm with a UV detector. The flow rate of mo- bile phase was 0.7 mL/min and the column temperature was 35 ℃ for the accurate determination of β-si- tosterol. The method has a good linearity within a range of 4.0× 10-7 ~2.0× 10-5 g(r=0. 999 9) under optimized conditions. The average recovery was 99.70%. The content of β-sitosterol in A. potaninii Korsh from Shaanxi Taibai Mountainous area was determined, and the average content of β-sitosterol was 0. 134%. This method is simple and accurate to be used for the evaluating quality of A. potaninii Korsh.
