摘要
目的探讨阿托伐他汀对氧化低密度脂蛋白(Ox-LDL)诱导大鼠胸大动脉平滑肌细胞(A7r5)增殖及缝隙连接蛋白43(Cx43)表达的影响。方法培养A7r5细胞株,将细胞分4组:对照组,Ox-LDL组,Ox-LDL+二甲基亚砜(DMSO)组及Ox-LDL+他汀组。MTS法细胞毒试验及流式细胞术检测细胞周期,从而检测A7r5的增殖活性;流式细胞术检测各组供体细胞Q3(发绿色荧光)与受体细胞Q4(双阴性)比值(Q3/Q4)的变化,从而分析GJ功能的变化;Western blot法检测A7r5细胞中Cx43表达。结果与对照组相比,Ox-LDL组及Ox-LDL+DMSO组A7r5细胞的增殖活性增加,S期细胞比率增高(P<0.05);与Ox-LDL组及Ox-LDL+DMSO组比较,Ox-LDL+他汀组二者均降低(P<0.05)。与对照组相比较,Ox-LDL组及Ox-LDL+DMSO组A7r5细胞的Q3/Q4比值增大,提示缝隙连接功能增强(P<0.05);与Ox-LDL组及Ox-LDL+DMSO组比较,Ox-LDL+他汀组Q3/Q4比值减小,缝隙连接功能显著降低(P<0.05)。与对照组相比,Ox-LDL组和Ox LDL+DMSO组A7r5细胞的Cx43表达增多(P<0.05);与Ox LDL组及Ox-LDL+DMSO组比较,Ox LDL+他汀组Cx43表达减少(P<0.05)。结论阿托伐他汀可以抑制氧化低密度脂蛋白诱导的A7r5细胞增殖及Cx43蛋白的表达。
Objective To study the effects of Atorvastatin on A7r5 cell proliferation and the expression of connexins 43 media- ted by OxLDL. Methods The A7r5 cell line was cultured, then the cells were divided into four groups, including Control group, Ox - LDL group, Ox - LDL + DMSO group and Ox - LDL + Statin group. The cellular proliferation activity was measured by MTS and Flow cy- tometry. Flow cytometry was used to detect the ratio of Q3 ( Calcein labeled cells) to Q4 ( double negative cells) for the analysis of the changes of the gap junction function. The expression of connexins 43 ( Cx43 ) was detected by Western blot. Results The A7r5 cell prolif- eration activity and the S - phase fraction detected in the Ox - LDL group and the Ox - LDL + DMSO group were increased obviously compared with those in the control group ( p 〈 0.05 ).
