摘要
目的检测喉癌及癌旁组织中蛋白酶活化受体1(PAR-1)、血管内皮生长因子(VEGF)、基质金属蛋白-2(MMP-2)的表达,及重组水蛭素(Recombinant hirudin)对喉癌细胞Hep-2中PAR-1、VEGF、MMP-2表达的影响,探讨PAR-1、VEGF、MMP-2在喉癌侵袭及转移中的作用及重组水蛭素的抗肿瘤机制。方法采用免疫组化方法检测喉癌组织及癌旁组织中PAR-1、VEGF、MMP-2的表达;用免疫细胞化学方法检测重组水蛭素共培养和不加入重组水蛭素培养的喉癌细胞中PAR-1、VEGF、MMP-2的表达。结果 PAR-1、VEGF、MMP-2在喉癌组织及有淋巴结转移的癌旁组织中呈高表达,差异有统计学意义;PAR-1、VEGF、MMP-2在重组水蛭素共培养和不加入重组水蛭素培养的表达差异有统计学意义(P<0.05)。结论证实PAR-1、VEGF、MMP-2参与了喉癌的侵袭及转移;重组水蛭素的抗肿瘤及抑制肿瘤转移作用可能与其降低喉癌细胞中PAR-1、VEGF、MMP-2表达有关。
Objective By detecting laryngeal cancer and cancer adjacent tissues of Protease Activated Receptorl ( PAR1 ), Vascular Endothelial Growth Factor (VEGF), Metal Matrix Protein -2 (MMP -2) high expression, discusses mechanisms of invasion and metastasis laryngeal cancer. By detecting PAR1, VEGF, MMP - 2 expression when Recombinant Hirudin in laryngeal cancer cell Hep -2,discusses the mechanism of Recombinant Hirudin against tumors. Methods Immunohistochemical method was used to detect the laryngeal cancer tissues and the tissue adjacent to carcinoma of PAR1, VEGF, MMP -2. Immunohistochemical method was detect Hirudin cultivation and un - Hirudin of laryngeal cancer ceils of PAR1, VEGF, MMP - 2. Result The expression of PARI, VEGF, MMP - 2 were higher in laryngeal carcinoma tissues. The difference was statistically significant. In Hirudin cultivation and un - Hirudin in laryngeal cancer cells,the expression of PAR1, VEGF, MMP - 2 exist significant difference, there are statistically significant. Conclusion Con- firmed PAR1, VEGF, MMP - 2 in laryngeal cancer invasion and metastasis process mutual synergy. Recombinant hirudin reduces the ex- pression of PAR1, VEGF, MMP -2 in laryngeal cancer ceils, thus antitumor and inhibition of tumor metastasis.
