树鼩转基因研究展望 被引量:1

Research Prospects of Transgenic Tree Shrew

摘要 树鼩是一种外形类似松鼠的小型哺乳动物,相对于其他小型哺乳动物在生理学、解剖学和遗传学上与灵长类有更高的相似度.树鼩作为动物模型已经取得了较多的研究进展,但目前树鼩并没有得到广泛的应用,主要原因之一是缺乏有效的遗传操作手段.常用的遗传操作细胞载体有1-细胞胚胎、能分化形成生殖细胞的多能干细胞和精原干细胞,其中精原干细胞是一种很有前景的遗传操作细胞载体,在精原干细胞上能通过各种分子生物学的方法对其遗传物质进行修饰.通过精原干细胞能够进行转基因树鼩的构建是一个重大的突破,为转基因树鼩构建提供一个平台,将促进树鼩基础生物学的研究,并为树鼩在医学生物学研究领域的应用提供重要基础. Tree shrew( Tupaia belangeri) is a kind of small mammal which looks like squirrel. Compared with other small mammals,tree shrews are more similar to primates in aspects of physiology,anatomy and genetics.As an animal model,it has made lots of progress. However,tree shrews are currently not widely used due to lack of effective method of genetic manipulation. Commonly used genetic operation cells include 1-cell embryo,pluripotent stem cells and spermatogonial stem cells. Spermatogonial stem cell is a kind of cell with good application prospects in genetic manipulation,which could be genetically modified through a variety of molecular biology methods. Constructing the transgenic tree shrew with the spermatogonial stem cells is a great breakthrough,which provides a platform for transgenic tree shrew,and promotes the basic research of tree shrew,as well as its application in medical biology.
