百草枯诱导急性肺损伤大鼠和肽素水平变化及甘草酸二铵对其影响 被引量:5

The dynamic changes of copeptin in rats with acute lung injury induced by paraquat and diammonium glycyrrhizinate intervention studies

摘要 目的探讨急性肺损伤(ALI)大鼠和肽素(copeptin)水平的变化情况,进一步探究甘草酸二铵(diammonion glycyrrhizinate,DG)干预对其表达情况的影响。方法以百草枯(paraquat,PQ)建立ALl大鼠模型,将150只sD大鼠按照随机数字表法分为PQ染毒组、DG干预组和空白对照组(NS)。PQ组和DG组大鼠给予PQ100mg/kg一次性灌胃,NS组大鼠则给予等量生理盐水灌胃。DG组大鼠灌胃后立即给予DG50mg/kg腹腔注射,每24h重复给药1次。分别于12、24、48h处死动物留取标本。ELISA检测血清和肺组织中和肽素、炎症因子TNF—α和IL-10浓度,并测定肺组织湿/干质量(W/D)比值。结果在12、24、48h三个观察时段,PQ组血清和肺组织中和肽素、TNF-α和IL-10浓度均逐渐升高,肺组织W/D比值增高,差异有统计学意义(P均〈0.001)。而此三个观察时段的组间比较发现,DG干预组血清和肺组织中和肽素、TNF-α浓度降低,而IL-10浓度升高,肺组织W/D比值降低,差异有统计学意义(P均〈0.001)。结论和肽素水平可能与肺损伤的严重程度有关。DG对ALI有保护作用。 Objective To investigate the effects ofdiammonium glycyrrhizinate (DG) on the levels of copeptin,TNF-α and IL-10 in rats with acute lung injury(ALI) induced by paraquat (PQ).Methods The PQ (100 mg/kg)were administered via a gastric tube to make an animal model of ALl in rats. Then DG (50 mg/kg per 24 h)were administered immediately by intraperitoneal injection in the DG group. The ELISA assay was applied to measuring the levels of copeptin ,TNF-α and IL-10 in lungs and blood. And the wet to dry ratio of lungs were measured. Results The levels of serum and pulmonary copeptin, TNF-α and IL-10 in PQ group were higher than those in NS group, and the pulmonary wet to dry ratio were increased (P 〈 0.001).It demonstrated that the pulmonary edema was aggravating. While, the levels of serum and pulmonary copeptin and TNF-α in DG group were lower than those in PQ group. However, the levels of IL-10 in DG group were increased (P 〈 0.001).And pulmonary wet to dry ratio were lower(P 〈 0.001 ), it stated pulmonary edema were attenuated. Gonelusions Copeptin levels were probably associated with the severity of pulmonary edema. And DG had protective effect against acute lung injury.
