两种内参基因在鸡循环miRNA相对定量检测中的比较分析 被引量:1

摘要 选择5SrRNA和snRNAu6作为内参基因,比较分析其在鸡血清循环miRNA定量检测中的应用效果。结果显示,5SrRNA和snRNAu6两个内参基因在不同发育阶段血清中表达稳定,5SrRNA的Cq值在24.4~25.0之间,snRNAu6的cq值在24.0-24.5之间,组间Cq值的变化幅度小于0.6;同一测定miRNA的组内Cq值变异也较小。两种内参基因条件下,3个测定循环miRNA的表达变化规律均一致。表明5SrRNA和snRNAu6是适于鸡血清循环miRNA定量检测的内参基因。 Two control genes,SS rRNA and snRNA U6,were selected to compare the effects in chicken (G. gallus) serum circulating miRNA real-time quantitative PCR (RT-qPCR) tests. The results showed that expression levels of 5 S rRNA and snRNA U6 were relatively stable between different groups, with 24.4 to 25.0 Cq values for 5 S rRNA and 24.0 to 24.5 Cq values for snRNA U6. The within-group Cq values for each tested miRNA also had less variation. The tested three miRNAs had the same expression tendency using either 5 S rRNA or snRNA U6. The results indicated that 5 S rRNA and snRNA U6 were suitable control genes for chicken serum circulating miRNA RT-qPCR test.
