摘要
选择6-BA、NAA、培养基种类和有机添加物种类等4个因素进行L16(4^4)正交试验,研究各因素对大苞鞘石斛原球茎分化的影响。结果表明:将无菌播种50d已突破种皮的原球茎转接于含3g/L花宝1号+2g/L蛋白胨+0.2mg/LNAA+10%马铃薯提取液+20g/L蔗糖(pH5.6)的培养基上培养70d,分化效果最好。6-BA对原球茎分化影响最为显著。
Four factors such as 6-BA, NAA, different culture medium and organic additives with 4 levels were selected to study the effects of different factors on the protocorm differentiation of Dendrobium wardianurn by orthogonal test. The results showed that the protocorm obtained by sterile culture for 50 d inoculated in the differentiation medium of Hyponex No. 1 3 g/L + peptone 2 g/L + NAA 0.2 mg/L + potato extract 10% + sugar 20 g/L(pHS. 6 ) for 70 d could get the best differentiation effect. 6-BA had the most significant effect on the differentiation of protocorm.
