摘要
本研究探讨头孢克肟对健康SD大鼠甲状腺功能的影响。采用SPF级雄性SD大鼠40只,随机分为正常组、对照组、头孢克肟高、低剂量组各10只。除正常组外,分别给予生理盐水、头孢克肟18 mg/kg、36 mg/kg灌胃,每日1次,连续4周。给药结束1周后,取血及甲状腺,ELISA法测定血清FT3、FT4、TSH、rT3,HE染色、光镜观察甲状腺组织病理变化。ELISA法结果显示,与正常组和对照组比较,头孢克肟高、低剂量组血清FT3、FT4、rT3水平均降低,差异有统计学意义(p<0.05),正常组与对照组之间对比无统计学意义(p>0.05)。HE染色结果显示,头孢克肟高、低剂量组甲状腺滤泡大小不均匀,滤泡明显萎缩,滤泡间可见大量淋巴细胞。本研究结果提示,头孢克肟可导致健康SD大鼠甲状腺功能减退。
The aim of this study was to investigate the effect of cefixime on thyroid function in healthy SD rats. 40 SPF male SD rats were randomly and evenly divided into normal group, control group, eefixime high and low dose groups. Except for the normal group, other 3 groups were orally given saline, cefixime with dose of 18 mg/kg and 36 mg/kg once daily, continuous 4 weeks. After administration of 1 week, we acquired blood and thyroids. Assay FT3, FT4, TSH and rT3 of serum with ELISA, we observed the pathological changes of thyroid under light microscopy after HE staining. ELISA assay showed that, compared with normal group and control group, the levels of serum FT3, FT4, rT3 in high and low cefixime dose groups were lower. The difference was statistically signifieant (p 〈0.05), while there was no significant difference between normal group and control group (p 〉0.05). HE staining showed that the thyroid folliculars were of non-uniform size, obviously atrophied, as well as a large number of lymphocytes between folliculars in cefixime high and low dose groups. The results of this study suggested that cefixime may cause hypothyroidism in health SD rats.
