参麦注射液对肺腺癌耐药细胞株的逆转作用 被引量:3

The Reversion of Drug Resistance in Lung Adenocarcinoma Cells Treated by Shenmai Injection

摘要 本研究探讨了参麦注射液对人肺腺癌耐药细胞株A549/DTX(多西他赛)的耐药逆转作用,并初步分析其作用机制。以MTT法检测参麦注射液、多西他赛及联合应用对耐药细胞株的增殖抑制率,并计算耐药逆转倍数。用流式细胞术测定参麦注射液联合多西他赛对A549/DTX细胞株的促凋亡作用;用Western blotting分析参麦注射液对A549/DTX细胞株的凋亡相关蛋白Bax和Bcl-2表达水平的影响。研究表明,参麦注射液增加耐药细胞株对DTX的敏感性,耐药逆转倍数为3.92,并明显增强DTX对A549/DTX细胞株的促凋亡作用。同时抑制了Bcl-2和P-gp蛋白表达,与对照组相比差异显著(p<0.05)。故参麦注射液能部分逆转A549/DTX细胞株耐药性,其作用通过诱导细胞凋亡及降低P-gp表达实现。 This study investigated the reversal effect of Shenmai injection on drug-resistant lung adenocarcinoma cell line A549/DTX (Docetaxel), and preliminarily analyzed its functionary mechanism. It used MTT assay to measure the proliferation inhibition rate of drug-resistent cell line with the effect of Shenmai injection, DTX, and both of them, calculating its reversing multiple. It also used flow cytometry to determine the promotion of apoptosis with the combined effect of Shenmai injection and Docetaxel. It applied Western blotting to analyze the influence caused by Shenmai injection on Bax and Bcl-2 (proteins related to the apoptosis of A549/DTX cell line) about their expression levels. The study showed that Shenmai injection increased the sensitivity of drug-resistant cell line to DTX, of which the reversal fold was 3.92. It significantly enhanced DTX's effect on the apoptosis of A549/DTX cell line. Moreover, the expression of Bcl-2 and P-gp was decreased compared with the control group (p 〈0.05). Therefore, Shenmai injection could partly reverse the drug resistance of A549/DTX cell line. Its mechanism was realized by apoptosis as well as inhibiting the expression of P-gp.
