二陈汤对脂代谢紊乱小鼠Cav-1和PI3K/AKT表达的影响 被引量:13

Effects of Erchen Decoction on the expressions of Cav-1 and PI3K/AKT in mice model of lipid metabolism disorder

摘要 目的:研究化痰方(二陈汤)对脂代谢紊乱C57BL/6小鼠肝组织中小窝蛋白-1(Cav-1)和磷脂酰肌醇3-激酶(PI3K)/蛋白激酶B(AKT)表达的影响。方法:选取24只SPF级雄性C57BL/6小鼠,用高脂饲料喂养10周建立脂代谢紊乱模型,于第4、10周检测其血脂含量,后随机分为3组:模型组、化痰组及降脂组,每组8只,另选8只小鼠为对照组。药物干预4周后,采用Real-time PCR检测肝组织中Cav-1、PI3K和AKT基因的表达。结果:造模后小鼠血脂[甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)]含量均升高,肝组织中Cav-1和AKT表达减少,PI3K表达增加;二陈汤使得模型小鼠血脂含量均有所降低,Cav-1和AKT表达得到回升,而PI3K表达持续增加。结论:化痰方(二陈汤)对脂代谢紊乱小鼠Cav-1/PI3K/AKT信号具有调控作用。 Objective:To observe the effects of Huatan Formula(Erchen Decoction) on the expressions of Caveolin-1(Cav-1) and PI3K/AKT in the liver tissue of C57BL/6 mice model ofdyslipidemia.Methods:Twenty-four SPF grade C57BL/6mice were fed with high-fat diet for 10 weeks to establish the mice model of dyslipidemia.The level of blood lipid was detected at the fourth and tenth weeks after modelling.Then 24 mice were randomly divided into three groups(n=8) including model group,Huatan group and Jiangzhi group.The control group was composed of 8 normal mice.After four weeks of drug administration,the mRNA expressions of Cav-1,PI3 K and AKT in the liver were detected by the method of Real-time PCR.Results:The administration of high-fat diet significantly enhanced the levels of blood lipid including TG,TC,HDL-C and LDL-C when compared with the control group,which were reversed by the administration of Erchen Decoction.Meanwhile,the administration of high-fat diet significantly decreased the expressions of Cav-1 and AKT in the liver and increased the expression of PI3 K.But,the expressions of Cav-1 and AKT in the liver were enhanced by the administration of Erchen Decoction.Additionally,Erchen Decoction had not yet showed obvious effects on the increasing PI3 K.Conclusion:Erchen Decoction,a representative for Huatan Formula,has a regulatory effect on Cav-1/PI3K/AKT signaling pathway in mice model ofdyslipidemia.
